Abstract


Background Psoriasis （OMIM#177900） is a common, chronic and recurrent, inflammatory skin disease. The pathogenesis of psoriasis remains unclear. Now most people tend to think it is a complex disease and caused by the interaction between genetics, genetical and environmental factors. Psoriasis vulgaris （PV） is the most frequent clinical type. We found a distinguished correlation of rs9304742 polymorphism in ZNF816A gene cluster with psoriasis vulgaris in Chinese Han Population by psoriasis genome-wide association study （GWAS）.Objective Based on stratifying analysis of the gender, age of onset, family history and clinical types, the study compared the genotype distribution of rs9304742 polymorphism within ZNF816A gene with clinical features of psoriasis vulgaris in the Chinese Han population in order to further explore the genetics pathogenesis.Methods We analyzed the distributions of genotype and allele of rs9304742 polymorphism within ZNF816A gene in 7,318 cases and 11,290 normal controls, which derived from the psoriasis genome-wide association study （GWAS）. The appropriate application of social science statistical software package SPSS 15.0 was used for statistical analysis of data.Results 1.The distribution of alleles and genotypes for rs9304742 polymorphism within ZNF816A gene were significant differences between cases and controls (1.80×10-8, 8.32×10-8, respectively). cases were stratified from the gender, age of onset, family history and clinical type, there were also significant differences between clinical subgroups and controls in distribution of allele and genotype （all P<0.01）. 2. The distribution of alleles and genotypes for rs9304742 polymorphism within ZNF816A gene weren’t significant differences between male cases and female cases groups （χ2=0.63,P=0.43;χ2=0.88,P=0.65; respectively）. 3. There weren’t significant differences between the pediatric onset psoriasis patients and the adult onset psoriasis patients in distribution of allele and genotype of rs9304742 （χ2=1.12,P=0.30;χ2=1.11,P=0.57; respectively）. 4. The distribution of alleles and genotypes for rs9304742 polymorphism within ZNF816A gene were also no significant differences between familial cases and sporadic cases groups （χ2=2.37 , P=0.13 ;χ2=4.43 , P=0.11; respectively）. 5. The difference in frequencies distribution of allele and genotype for rs9304742 between the guttate and plaque cases did not reach the statistical significance （χ2=0.004,P=0.95;χ2=0.47,P=0.79; respectively）.Conclusions 1.The rs9304742 polymorphism within ZNF816A gene is associated with the susceptibility of psoriasis vulgaris in the Chinese Han population. 2. The rs9304742 polymorphism within ZNF816A gene is not associated with gender, age of onset, family history and clincal type of psoriasis.
