Experimental Study of Recombinant Human Endostatin Combined with Chemotherapy of NVB-Cisplatin on Human Breast Cancer Xenografts in Nude Mice

Objective: To evaluate the inhibitory effect of recombinant human endostatin (rh-Endostatin, rh-ES) combined with chemotherapy of NP treatment on tumor growth and antiagiogenesis in nude mice after xenograft of MCF-7 cell line.Methods: 60 nude mice models with human breast carcinoma were divided randomly into group NVB-Cisplatin (A), group rh-ES (B), group NVB-Cisplatin + rh-ES (C) and control group (D), each group had 15 nude mice. Group A was given NP treatment one cycle. Group B was given rh-ES two cycles. Group C was given NP treatment one cycle combined with rh-ES two cycles. Group D was given normal saline solution respectively. NVB 10 mg.kg-1, d1,8;DDP 10 mg.kg-1, d3-5. Rh-ES was used two cycles, each cycle for 14 days, two cycles interval 7 days. After the second cycle, ten mice were put to death in each group. Tumor inhibition rate (TIR) and growth curve of each group was calculated and plotted. The vascular endothelial growth factor (VEGF) and microvascular density (MVD) expression were detected by immunohistochemistry. The leftover mice of each group were raised utill they dead. The survival time of leftover mice were observed. The tumor volume, the expression of VEGF and MVD, the survival time were all analysed by statistics.Result: TIR of groups A, B and C were 61.8%, 64.79% and 88.95%, respectively. The TIR of group C was significantly higher than other two groups. Compared with VEGF expression, group C was significantly lower than other two groups (both P were 0.001). The MVD in each group were 43.50±8.17、29.20±5.90、11.60±1.77 and 53.20±5.78, respectively. Group C was also significantly lower than other groups (P were 0.000, 0.006 and 0.000). The survival time in each group were 116.00±14.71 d、134.40±14.15 d、154.40±24.76 d and 110.60±9.02 d. The survival time of group C was longer than groups A and D ( P were 0.003 and 0.002).Conclusion: Rh-ES combined with chemotherapy of NP have synergistic effect, they can obviously inhibit tumor growth and angiogenesis. They also can obviously prolong the survival time of nude mice.
