Abstract


Objective To investigate the Snail regulation of epithelial -mesenchymal transition in colorectal cancer invasion . The role and significance of the transfer . Method detected 40 cases of colorectal carcinoma and adjacent tissues in the Snail, E-cadherin expression by immunohistochemistry . The 2 siRNA interference Snail genes after colorectal cancer SW480 cells were divided into the control group . Empty plasmid group and siRNA-Snail transfected group , RT-PCR were used to detect colorectal SW480 cells of Snail , E- cadherin mRNA expression ; the Transwell chambers stromal invasion assay and heterogeneity adhesion assay were detected colorectal SW480 cell invasion and heterogeneous adhesion ability . Results 1 colorectal tissue of Snail , E - cadherin positive expression rate of 57.5% , respectively . 27.5% , cancer tissue cells in the positive expression rate of 10% . 80%; these two proteins in colorectal cancer tissues and paraneoplastic organizations cells were significant differences ( P lt; 0.05 ) were associated with colorectal cancer differentiation and lymph node metastasis correlation ( P lt ; 0.05 ), and gender . Age related to sex ( P gt ; 0.05) , and the expression of the two was a negative correlation ( P lt; 0.05 ) . 2 RT-PCR to detect the SW480 cells of Snail , E- cadherin mRNA results show that compared with the control group , Snail, E- cadherin mRNA expression in empty plasmid group was not statistically significant (P gt; 0.05), transfected with group statistically significant ( P lt; 0.05 ) . The 3 matrix invasion assay and heterogeneous adhesion Experimental results show that compared with the control group , the transfected group junction cancer SW480 cell invasion force and heterogeneity adhesion decreased, the difference was statistically significant ( P lt; 0.05 ) , empty plasmid group showed no statistical significance ( P gt ; 0.05 ) . Conclusion Snail regulation of epithelial -mesenchymal transition is closely related to the invasion and metastasis of colorectal cancer , Snail, E-cadherin may serve as indicators of the invasion and metastasis of colorectal cancer .
