Background: Oxidative stress can induce lens epithelial cell apoptosis the antioxidant epigallocatechin stones catechin gallate has anti-apoptotic effects, but the specific mechanism of action remains unclear. Objective: To observe the Fas protein and hydrogen peroxide-induced cataracts in the lens epithelial cell apoptosis and stones catechin epigallocatechin gallate Fas protein and apoptosis of lens epithelial Methods: transparent crystals of healthy adult rabbits were randomly divided into three groups: control group, added only DMEM culture medium; peroxide group and blank control group 3% hydrogen peroxide, hydrogen peroxide in a final concentration of 0.3 mmol / L; after roughly stones catechin epigallocatechin gallate group hydrogen peroxide group stones catechin epigallocatechin gallate, epigallocatechin stones catechin gallate a final concentration of 0.1 mmol / L, were cultured in vitro for 72 h observation lens turbidity light microscope observation of lens tissue morphological changes detected lens antioxidant defense system superoxide dismutase (SOD) and catalase (CAT) content and lipid peroxidation end product malondialdehyde aldehyde (MDA) content, using the terminal deoxynucleotidyl transferase nick end labeling in situ cell assay (TUNEL) detection of lens epithelial cell apoptosis rate, the immunohistochemical assay lens epithelial cell expression of Fas protein. Compared with the control group, H 2 O 2 group SOD, CAT levels were significantly reduced MDA levels were significantly higher, with H 2 O 2 group of EGCG group SOD, CAT levels significantly increased MDA levels were significantly reduced; lens epithelial cell apoptosis rate, H 2 O 2 < / sub> group (96.786 ± 0.9465)% was significantly higher than the control group (3.422 ± .8365)%; of EGCG group (27.086 ± 0.2754)% significantly the below H 2 O 2 group; Fas protein expression was H 2 O 2 group (60.976 ± 0.4709)% was significantly higher than the control group (9.674 ± 0.6082)%, EGCG group (19.564 ± 0.4296)% was significantly lower than the H 2 O 2 group. The above differences were statistically significant (P lt; 0.05). Conclusion: increase the expression of Fas protein may be one of the mechanisms of hydrogen peroxide induced lens epithelial cell apoptosis, and antioxidant EGCG can down-regulate the expression of Fas protein and reduce apoptosis of lens epithelial cells.
