Objective To observe and compare the therapeutic effects of curcumin and its derivatives on experimental nonalcoholic steatohepatitis (NASH). (1) rat NASH model: 60 male Sprague-Dawley rats were completely randomized principles grouped, divided into normal group (A), model group (B), saline group (C), turmeric Su gavage group (D), the curcumin derivatives group (E). 12 in each group. A group were fed a normal diet and tap water. Groups B, C, D, E in the limbs, the medial subcutaneous injection of 40% CCl 4 : paraffin oil solution at a dose of 3 ml / kg, twice a week, the first dose of doubling; given at the same time containing 79.5 % cornmeal, 20% lard, 0.5% cholesterol, high-fat diet. Group A and group B rats were sacrificed after 6 weeks 3 each, serum and pathological examination of observed fatty liver model-making effect. (2) the treatment of NASH and its mechanism of testing curcumin and its derivatives: modeling for six weeks, A, B groups of all rats were sacrificed. , D group given curcumin gavage doses of 50mg/kg 2ml / times, day 1; C group, E group were given tail vein injection of saline and curcumin derivatives, the dose of 50mg/kg daily 1 times; continuous treatment for two weeks. Take the blood and liver tissue after the end of treatment, liver index, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), total cholesterol (TCH) content detection. HE and Oil Red O staining of liver tissue was observed histopathological changes of the liver tissue. RT-PCR assay of liver tissue TNF-α, NF-κB, FAS, HMG-CoA reductase mRNA transcription, Western blot detection of TNF-α and NF-κB protein expression. Results (1) the end of the experiment, the body weight of rats were increased. In addition to the normal group, all rat liver index of more than 5%, to the requirements of the diagnosis of fatty liver model group, liver index was significantly higher than the normal group, the difference was statistically significant (P lt; 0.05). HE staining showed that the model rats serious injury pathological changes, general fatty degeneration of the liver cells, inflammation, necrosis and bleeding obvious, a small number of rat hepatic portal area visible fibrous tissue hyperplasia. Model group, the degree of steatosis and inflammation grade, almost at the highest level, but to a lesser extent fibrosis. Compared with the normal group, the model rats serum ALT, AST, TCH water was significantly higher (P lt; 0.05); mRNA transcription levels of TNF-α, NF-κB, FAS and HMG-CoA reductase four genes significantly elevated (RQ gt; 1); both NF-κB, TNF-α protein expression was significantly increased. (2) after saline, curcumin and curcumin derivatives in the treatment of, all rat liver index were decreased compared with the model group, the difference was statistically significant (P lt; 0.05). Curcumin and its derivatives in serum ALT, AST, TCH than the saline group, the difference was statistically significant (P lt; 0.05). In serum ALT, AST indicators, the level of the derivatives group was significantly lower than the gavage group, the difference was statistically significant (P lt; 0.05). No significant changes in levels of triglycerides (TG), before and after treatment, the difference was not statistically significance (P gt; 0.05). HE and Oil Red O staining of liver steatosis of the curcumin derivatives rats compared with the model group significantly reduced inflammation grade also improved markedly. Compared with normal saline group of curcumin derivatives fibrosis light, the difference was statistically significant (P lt; 0.05). Gavage group compared with model group degree of steatosis, inflammation grade and fibrosis had no significant statistical significance (P gt; 0.05). RT-PCR results suggest that by curcumin and its derivatives for the treatment of liver tissue TNF-α, NF-κB, HMG-CoA reductase and FAS mRNA transcription level was significantly lowered, but less obvious derivatives group curcumin with derivatives group, the difference was statistically significant (P lt; 0.05). Western blot visible compared with the saline group, curcumin gavage group and derivatives group NFκB and TNFα protein expression was significantly decreased, the difference was statistically significant (p lt; 0.05). Saline group with model group, the difference was not statistically significant (p gt; 0.05). Conclusions relative to traditional curcumin, curcumin derivatives has a more pronounced anti-inflammatory and lipid-lowering effects of experimental NASH rats.
