Objective: osteoporosis (Osteoporosis, OP) is based on per unit volume of bone tissue reduced the fine structure of the bone tissue damage, bone fragility characterized by a systematic, systemic bone disease. OP two cells are osteoblasts (osteoblast, OB) and osteoclasts (osteoclast OC). This paper studies the pulsed electromagnetic fields (pulsed electromagnetic fields, PEMFs) on the OC, OB and their precursor cells. PEMFs (?) Taiwan treatment of osteoporosis mechanism discussed to provide a basis for clinical PEMFs applications. Methods: a pulsed electromagnetic fields on osteoclast precursor cells and osteoblast precursor cells: an in vitro PEMFs role: separation of 8w-year-old female SD rat femur bone marrow cells, cultured in vitro accept PEMFs role. 2, the in vivo binding PEMFs role vitro: the 8w age female SD rats in vivo in ovariectomized accept PEMFs role 8W separation of the femur bone marrow cells were cultured in vitro, the PEMFs role continue to accept or not accept. Colony formation were cultured and observed the CFU-F (fibroblast colony forming units fibroblast colony-forming units) and CFU-GM (granulocyte / macrophage colony Forming unit, and granulocyte / macrophage colony-forming units). Second, the impact of pulsed electromagnetic fields on osteoclast: 8w age live female SD rats were ovariectomized and accept PEMFs role 8W separation of the femur bone marrow cells were cultured in vitro, continue to accept or not to accept PEMFs role. Were induced cultured osteoclasts, and tartrate-resistant acid phosphatase (tartrate-resistant acid phosphatase, TRAP) detection, integrin avβ3, NF-kappa B receptor activator (receptor activator of NF-kappaB, RANK) cathepsin K (Cathepsin K) immunocytochemistry staining. Pulsed electromagnetic fields on osteoblasts: separation of the 6w-year-old female SD rat femur and tibia bone marrow cells, induction into the process of bone cells in vitro PEMFs role of NF-kappa B receptor activating factor ligand (receptor activator of NF-kappa B ligand, RANKL), ALP (alkaline phosphatase, ALP) detection. : 1 isolated PEMFs role, the number of CFU-GM cloning reduce, increase in the number of CFU-F clone; 2, in vivo combination isolated PEMFs role, the dose groups CFU-F were increased, and each group of CFU-GM There was no significant difference. PEMFs after OC number was significantly reduced;, immunocytochemistry staining showed that OC is also significantly reduced. The PEMFs role, OB number increased significantly OB differentiation in vitro was markedly increased. Conclusion: isolated PEMFs role PEMFs OC precursor cells inhibition. However, the in vivo binding isolated PEMFs role, no inhibition of OC precursor cells. 2, from the body PEMFs and in vivo binding in vitro PEMFs effect, both to promote the proliferation of precursor cells of the OB. PEMFs on the differentiation and function of the OC inhibition, can inhibit the biological activity of OC bone resorption. PEMFs OB differentiation and function can promote, promote OB activity.
