Abstract


The purpose of detection of human breast cancer cells Rab27A positioning express Rab27A in vitro and in vivo studies with breast cancer cell invasion and metastasis . Method 1 , using SYBR GREEN real-time semi - quantitative RT-PCR technology to detect Rab27A four human breast cancer cell line MCF -7 and MDA - MB -231 , MDA - MB -435 and MDA-MB-435HM mRNA applications Western-blot to detect Rab27AmRNA in the four human breast cancer cells MCF - 7 , MDA - MB -231 , MDA - MB -435 and MDA-MB-435HM protein expression . , Immunofluorescence (immunofluorescence) and laser confocal scanning imaging techniques (confocal) positioning Rab27A protein in breast cancer cells . 3 fat pads of nude mice breast (mammary fat pad, MFP) inoculated with human breast cancer cells , to explore Rab27A overexpression of lung metastasis of orthotopic tumor growth . Results 1 Rab27A mRNA and protein in the human breast cancer cell line MCF -7 , elevated levels of expression in MDA - MB -231 , MDA - MB -435 and MDA-MB-435HM with enhanced cell invasion and metastasis . 2 , in human breast cancer cell line MCF -7 and MDA - MB -231 and MDA-MB-435 , Rab27A diffuse distribution in the cytoplasm , and the latter two visible Rab27A protein in the perinuclear aggregation . 3, Rab27A high expression cell clone group increased rate of lung metastases in nude mice , MDA - MB -231 group by 10 % to 50 % ( P (0.01 ) , MDA - MB -435 group by 60% to 90% of (P < 0.05) , the average number of each intrapulmonary metastases also significantly increased the group in MDA-MB-231 and MDA -MB ??- 435 groups , respectively 5 and 2.5 times the control group ( P <0.01 ) , the volume of lung metastases increases, the and visible the multiple metastases fusion phenomenon . conclusions elevated of Rab27A expression levels with enhanced invasion and metastasis of breast cancer cells , orthotopic tumor growth and formation of lung metastases in vivo effects of , on the Rab27A expression may be related to the invasion and metastasis of breast cancer cells .
