Abstract


Objectives:To study the expression of CD59 on T cells by raising tumor model in murine.We study the effect of T cells and the changes of the signal transduction in T cells with CD59mAb and peptide seals specific to CD59,and investigate the mechanism of them.Our study and research will give a new way in tumor therapy.Methods:BALB/C rats were divided evenly into 3 groups by random.Tumor models were developed by HeLa cells or HeLa cells with CD59-transfected gene in groupⅡ,Ⅲ,and GroupⅠwas treated as normal control.After the establishment of the model,the rats were executed in the 20 days to do the following experiment:a.segregate blood serum to detect the level of complement C3,C4;b.do HE stain of tumoral tissue to observe the pathological change;c.detect the expressions of the tumoral tissue in groupⅡ,Ⅲ;d.detect the expressions of CD59 on T cells from thymus by FIH and EIH;e.withCD59mAb and peptide seals specific to CD59 to the T cells,detect lymphocyte proliferation in thymus by lymphocyte transformation test(MTT assay);f.Laser scanning confocal microscope imaging of the calcium fluorescent indicator dye Fluo-3/AM was used to determine the kinetic changes of [Ca2+]in T cell endochylema.CD4+CD25+ T cells were determined by flow cytometer.Results:The tumoral rat model was successfully developed by HeLa cells and HeLa cells with CD59-transfected gene.After development of the model,grouplII was easier than groupⅡin raising tumoral model and the blood compiement C3,C4 level in groupⅡ,Ⅲwas obviously lower than that of control group(P<0.01).The expression of CD59 on T cells in tumor organism reduced by FIH and EIH(P<0.05).The proliferation of lymphocyte in groupⅡwas evidently reduced comparing with the control group(P<0.01).The degree of the [Ca2+]i and the expression of CD4+CD25+ T cells all depressed comparing with the control group(P<0.01).The peptide seals specific to CD59 can increase[Ca2+]and inhibit the proliferation of T cells.Conclusions:The expression of CD59 on T cells can reduce in murine.CD59mAb can increase the[Ca2+]and make the resting T cells into CD4+CD25+T cells.The peptide seals specific to CD59 can block the CD59 of T cells.
