Abstract


Phthalate esters(phthalate esters PAEs)is the one of artificially synthesized organic compounds that has the biggest production and wide application,and it is a global environmental pollutant,widely existing in the air,water,soil and organism.Benzyl butyl phthalate(C19H20O4) is a sort of phthalateesters PAEs,which functions as plasticizer, softener,carrier and additive.It is classified as a important environment monitoring target by the world because of its anti-retrogradation,various pollution sources,high level of pollution,extensive toxicity,and causing various degree of danger to human health.This research is oriented on genotoxicity and oxidative damage of Benzyl butyl phthalate,and studies its molecular mechanism,which is of great significance for a full understanding of the genetic toxicity of BBP,and provides a certain experimental data for the study of the genetic toxicity of BBP and oxidative damage.My research is objected on mouse liver cell and Hela cell to study genetic toxicity of BBP by DNA-protein cross-linking experiments to explore the genetic toxicity,and study its oxidative-damage effect by means of detecting activity of SOD and MDA occupancy. Experimental results are as follows:1.BBP induced DNA-protein cross-linking effectUsing mouse liver cell and Hela cell as research material,the DPC induced by BBP was studied by application of SDS-KCl precipitate.Results showed that BBP could urge a production of DPC in mouse liver cell and Hela cell.Among them,the DPC effect at low concentrations is not obviously,only in the medium and high concentrations can cause significant DPC effect.2.BBP induced oxidative damage effectUsing intraperitoneal injection to mice in different concentration doses of BBP, determination SOD activity and MDA content of liver tissue cells,the effect of oxidative damage induced by BBP on mouse liver cells was detected.The results showed:SOD activity in liver cells has a declining trend with the increased concentration of BBP.when BBP exposure the concentration of 125mg/kg,the decreased Of the activity of SOD in the mouse liver cells ont obviously,there is no significant difference(P>0.05);and while BBP exposure in the concentration of 250mg/kg,500mg/kg and 1000 mg/kg,there is a very significant difference(P<0.01).With the increased concentration of BBP exposure, MDA content of liver cells in mice also increased,and compared with the control group, the MDA content of liver cells in mice increased one by one,and this increase has significant difference in concentration of 250mg/kg.This result showed the opposite trend coincides with the SOD activity effect of mouse liver cells caused by BBP.
