Abstract


Objective To study the Toll-like receptors (Toll-like receptor, TLR) 2.4mRNA leprosy patients in peripheral blood mononuclear cells and its related sites (TLR2-597T/C.TLR4-896A/G) gene single nucleotide polymorphisms (single nucleotide polymorphisms, SNP), to explore its contact with leprosy patients as well as in immune mechanism of leprosy infection. Methods China Guizhou Bijie leprosy cases 150 cases, of which 103 cases of tumor type, tuberculosis-like type 47 cases, 43 cases of familial aggregation, non-familial clustering of those 107 cases were detected by RT-PCR in peripheral blood mononuclear leprosy patients cells TLR2.4mRNA expression levels; using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of each sample TLR2 597T / C and TLR4896A / G locus genotype, genotype comparison leprosy and normal group as well as the percentage distribution of leprosy cases in each group were genotype possession ratios. SPSS16.0 software using ANOVA and X2 test for each set of data for statistical analysis. Results Leprosy group: TLR2mRNA 0.56 ± 0.06, control group: 0.22 ± 0.10, leprosy group: TLR4mRNA 0.34 ± 0.04, control group: 0.21 ± 0.07, TLR2.4mRNA leprosy patients in peripheral blood mononuclear cells were significantly higher expression (P lt; 0.01). Lepromatous leprosy group: TLR2mRNA 0.62 ± 0.07, TLR4mRNA0.40 ± 0.05; tuberculoid group: TLR2mRNA 0.43 ± 0.11, TLR4mRNA0.21 ± 0.07; control group: TLR2mRNA 0.22 ± 0.10, TLR4mRNA 0.21 ± 0.07, TLR2mRNA in tumor type and tuberculoid leprosy expression were higher (P lt; 0.05), lepromatous leprosy and tuberculosis-like expression of type group compared with no significant difference (P gt; 0.05); TLR4mRNA expression in lepromatous leprosy significantly higher (P lt; 0.05), in tuberculoid leprosy expression compared with the control group no significant difference (P gt; 0.05), in lepromatous leprosy was significantly higher than the tuberculoid group (P lt; 0.05). Familial aggregation group: TLR2mRNA 0.46 ± 0.12, TLR4mRNA 0.27 ± 0.08; non-familial aggregation group: TLR2mRNA 0.60 ± 0.08, TLR4mRNA 0.37 ± 0.05; TLR2. 4mRNA in familial aggregation group and non-familial aggregation group expressed no significant difference (P gt; 0.05). TLR2 (597T / C) TT, TC, CC genotype of leprosy patients and controls in the distribution of frequencies were 43.3%, 38.7%, 18% and 48.0%, 42.0%, 10.0%, no significant difference (P gt; 0.05); lepromatous and tuberculoid TT, TC, CC genotype percentage were 41.7%, 38.8%, 19.5% and 46.8%, 38.3%, 14.9%, the difference was not statistically significant (P gt; 0.05); familial aggregation and non-familial clustering of TT, TC, CC genotype percentage were 48.8%, 37.2%, 14.0% and 41.1%, 39.3%, 19.6%, the difference was not statistically significant (P gt; 0.05). All of the subjects did not detect TLR4 (896A / G) polymorphism genotypes were 896AA homozygous wild. Conclusion TLR2, 4mRNA leprosy patients in peripheral blood mononuclear cells was significantly increased, suggesting that TLR2, 4 may be leprae specificity recognition receptors, TLR4 lepromatous leprosy may also play an important role in the occurrence, TLR2, 4 in the onset and progression of leprosy may play a role. TLR2 (597T / C), TLR4 (896A / G) polymorphism and China Guizhou Bijie leprosy no correlation.
