The Study on Toxicology of Oxidation and Immunity of Rats Induced by PM2.5 Collected from Nickel-polluted Areas

Objective Through the comparative study of the acute toxic effects on the respiratory,immune and genital system of rats induced by ambient PM2.5 from different Nickel-polluted areas to explore the possible toxicology mechanism of PM2.5.Methods A nickel-polluted area and a control area were selected,the environment ambient quality of the two areas was monitored and the concentration of nickel in the collected PM2.5 was determined.Forty two male adult Wistar rats were randomly divided into three groups,including one saline control group and two experimental groups,the nickel-polluted area group and the control area group.in each,there were three subgroups.PM2.5 were administered to rats by intratracheal instillation at various doses(0、1.6、8.0、40.0mg/kg(BW)).After 24h exposure,The rats were sacrificed and studied on toxicological effects of their respiratory,immune and genital system.Results(1)There was no significant differences between the level of environment ambient quality in the two areas,while the concentration of nickel in PM2.5 in the nickel-polluted area was 48.75 times as that in the control area.(2)The results of toxicological study:①The phagocytosis rate of alveolar macrophage of the nickel-polluted area group were lower than that of the control area group,there was a significant difference in high dose;Well,similar condition also happened in the study of DNA damage by SCGE;while there were significant differences in the activities of ACP and AKP between the two experimental groups in middle dose,and LDH in high dose.②Lymphocyte stimulation index was only detected to be significantly lower in the nickel-polluted area group than the control area group in high dose.③Significant differences of the number of diploid cell and G0/G1 phase cell and PI between the two experimental groups were only found in middle dose;there were significant differences of the activities of T-AOC,SOD and MDA between the two experimental groups in high dose.Conclusions Both kinds of PM2.5 from the two different Nickel-polluted areas can induce oxidative damage and immune toxicitY in rats,it was more serious in nickel-polluted areas.nickel may play a key role in the toxicity of PM2.5 in the nickel-polluted area.
