Objective:To investigate the anti-atherosclerosis effects of Boschniaria rossica and its mechanism through experimental atherosclerosis model of animals, and to provide theory guidance for clinic exploitation of anti-atherosclerosis drugs. Methods:The experimental atherosclerosis model of rats were set up by feeding with atherogenic-diet and injecting vitamin D3(150,000U/kg by intramuscular injection in right lower limb once a week).Wistar male rats used in experiment were randomly divided into four groups:the normal group(normal diets);model group(atherogenic-diets);BR-treated group(atherogenie-diets +Boschniaria rossica); SI-treated group(atherogenic-diets+Simvastain).Boschniaria rossica and Simvastain were performed intragastric administration when atherogenic-diets were given, BR-treated group was performed intragastric administration of 160mg /kg.d Boschniaria rossica suspension,SI-treated group was performed intragastric administration of 9mg /kg.d Simvastain suspension, model group and the normal group were performed intragastric administration of the equal volume of drinking water, once a day,8 weeks in total. The level changes of Serum total cholesterol、triglyceride、low density lipoprotein cholesterol、high density lipoprotein cholesterol were measured after 8 weeks administration, the contents of malondialdehyde and the activity of superoxide dismutase in blood serum were detected. The experimental atherosclerosis model of rabbits were set up by feeding with atherogenic-diets.Wistar male rabbits used in experiment were randomly divided into four groups:the normal control group(normal diets);model group(atherogenic-diets); BR-treated group(atherogenic-diets +Boschniaria rossica);SI-treated group(atherogenic-diets+Simvastain).Boschniaria rossica and Simvastain were performed intragastric administration when atherogenic-diets were given,BR-treated group was performed intragastric administration of 80mg/kg.d Boschniaria rossica suspension,SI-treated group was performed intragastric administration of 5mg/kg.d Simvastain suspension,Model group and the normal group were performed intragastric administration of the equal volume of drinking water, once a day,8 weeks in total. The aortas of rabbits were separated, the pathological changes of aortas were observed under light microscope after HE staining after 8 weeks. Results:Compared with model group,BR could markedly decrease the total cholesterol、triglyceride、low density lipoprotein cholesterol in blood serum, which had a significant difference(P<0.01,P<0.01,P<0.05),BR could markedly increase high density lipoprotein in blood serum, which had a significant difference(P<0.01), BR could significantly reduced the contents of malondialdehyde in blood serum, which had a significant difference(P<0.01),BR could markedly enhanced the activity of superoxide dismutase in blood serum, which had a significant difference(P<0.01); BR could reduce endothelial cell engorgement of artery wall、arrangement derangement of smooth muscle and the histoleucocyte adhesion of atherosclerosis model rabbits. Conclusion:BR had antioxidant effect,It’ s possible mechanism could be attributed to regulating the lipid metabolism of rats, resisting the injury caused by oxidation free radical and preventing the injury of atherosclerosis artery endomembrane.
