Abstract


Ovarian cancer is one of the most common malignant tumors in women, and its mortality is the first.Ovarian conceals cavum pelvis deeply.On the pathogenetic early stage,deficiency of characteristic symptom and physical symptom makes the discovery and diagnosis difficult.About 70% of ovarian cancers enter its advanced stage when they are diagnosed.Compared with other malignant gynecology tumors,ovarian cancer is charactered with high-malignancy,early faraway-transferring and being removed difficulty. The 5 years survival rate of ovarian cancer is 30% or so.It has clinical sense to find influential factors and relatively mechanism of ovarian carcinomas biological behaviour,and to find method and index used in diagnosing early, detecting recurrence and evaluating prognosis.CyclinD1 is an important gene in the regulating-net of cell cycle.It plays a crucial role in the process of cell proliferation by participating the G1 stage of cell cycle.In recent years,experimental studies suggest CyclinD1 and its proteins are correlated with human tumors intimately. CyclinD1 is proved a new oncogene.Cyclooxygenase(COXs) are the rate-limiting enzymes that are neces- sary for converting arachidonic acid into prostaglandinH2.Two types have been described into COX-1 and COX-2.COX-2 belongs to inducible enzyme which is highly inducible by inflammatory stimuli,such as GF,cytokine and serum,etc.More recently,several in vitro and preclinical studies showed that COX-2 plays a role in the growth of ovarian cancer due to its ability to act as a tumor promoter.NOS is the major factor for the synthesis of NO.iNOS belongs to inducible NOS which is highly inducible by tumor cytokine stimuli,thus regulat the synthesis of NO.Moreover,NO can induce endothelial cell division,hyperplasy,and regulate neogenesis and blood flow of tumor vessel.More recently research showed that iNOS is important factor in promoting cell malignant transformation.Objective:The expressions of CyclinD1,COX-2 and iNOS in normal ovarian and epithelial ovarian tumors were detected to explore the new methods in research,prevention and therapy of ovarian carcinomas.Methods:23 specimens of ovarian carcinomas,20 specimens of benign ovarian tumors and 20 specimens of normal ovaries were tested by immuno- histochemical technical with antibody against CyclinD1,COX-2 and iNOS.Results:1.The positive rate of CyclinD1 expression in ovarian cancer was most highest.whereas(82.6%) there was no CyclinD1expression in ovarian benign tumor and normal ovarian tissue .The positive rate of CyclinD1 was significant difference between ovarian cancer and ovarian benign tumor(P<0.05).The positive rate of CyclinD1 was also significant diffe- rence between ovarian cancer and normal ovarian tissue(P<0.05).However,there was no difference between ovarian benign tumor and normal ovarian tissue in CyclinD1 expression(P>0.05).2.COX-2 expression in ovarian cancer was most highest. whereas(91.3 %) COX-2 expression in ovarian benign tumor and normal ovarian tissue was obviously lower.The positive rate of COX-2 was significant difference between ovarian cancer and ovarian benign tumor(P<0.05).The positive rate of COX-2 was also significant difference between ovarian cancer and normal ovarian tissue(P<0.05).However,there was no difference between ovarian benign tumor and normal ovarian tissue in COX-2 expression(P> 0.05).3.iNOS expression in ovarian cancer was most highest. whereas(73.9 %) iNOS expression in ovarian benign tumor and normal ovarian tissue was obviously lower.The positive rate of iNOS was significant difference between ovarian cancer and ovarian benign tumor(P<0.05).The positive rate of iNOS was also significant difference between ovarian cancer and normal ovarian tissue(P<0.05).However,there was no difference between ovarian benign tumor and normal ovarian tissue in iNOS expression(P> 0.05).Conclusion:The expression of cyclinD1,COX-2 and iNOS in ovarian carcinomas was significantly higher than that in benign ovarian tumors and normal ovaries,respectively. The overexpression of these markers may be involved in pathogenesis of ovarian carcinomas.
