Oral cancer (oral verrucous carcinoma, OVC) is a different oral squamous cell carcinoma, and has a unique pathological features and biological behavior of malignant disease, its pathogenesis is not clear. In this study, through the analysis of DNA methylation in the pre-test chips and whole genome expression differences in gene from selected target genes for further research. Explore OVC KLK KRT13 gene expression, to further explore the molecular mechanisms and therapeutic targets OVC occurred lay the foundation for real-time PCR and immunohistochemical techniques. Purpose: early oral the verrucous carcinoma DNA methylation chips and whole-genome chip differential expression of genes analyzed, preliminary verification chip accuracy, and explore KLK6 KRT13 gene in oral expression. Methods: 4 cases of oral verrucous carcinoma cancer tissue and corresponding normal mucosa of patients, cancer tissue, using real-time quantitative PCR validation KLK6 KRT13 genes in tissues, followed by immunohistochemical method in 19 cases of oral verrucous carcinoma, 26 cases of oral squamous cell carcinoma and 14 cases of normal oral mucosa KRT13 protein expression were detected by statistical analysis, KRT13 protein differentially expressed in different tissues. Results: Real-time PCR results showed that KLK6 Oral cancer, cancer adjacent normal tissue (method the paragraph does not say that the adjacent tissues,) in expression were decreased, but the difference was not statistically significant (P gt ; 0.05); KRT13 expression in the OVC lower than normal tissue and the difference was statistically significant (P lt; 0.05), no statistically significant difference in the expression in adjacent normal, cancer and paraneoplastic organizations ( P gt; 0.05). Positive rate was 57.89% (11/19), 26.92% (7/26) in oral squamous cell carcinoma, normal mucosal tissue 92.86% (13/14), in the three groups in the differential expression of KRT13 protein expression in oral cancer statistically significant (P lt; 0.05); KRT13 positive rate of outer-type OVC organization 87.50% (7/8) the cyst / infiltrating 36.36% (4/11), oral scores of scales carcinoma was 54.55% (6/11), poorly differentiated squamous cell carcinoma of the oral cavity is 6.67% (1/16); OVC exogenous cysts / infiltrating oral squamous cell carcinoma poorly differentiated tissue expression The differences were statistically significant (P lt; 0.05), the expression differences in well-differentiated oral squamous cell carcinoma in poorly differentiated tissue also statistically significant (P lt; 0.05), moderately differentiated oral squamous cell carcinoma and oral verrucous cancer exogenous oral squamous cell carcinoma, poorly differentiated OVC cyst / infiltration expression differences without statistical significance (P gt; 0.05). Conclusion: 1.KRT 13mRNA OVC low expression, consistent with the results of preliminary gene chip, protein expression is also lower than the normal mucosa, KRT may play some role in the development of 13 OVC no difference; KLK6 mRNA in oral expression and normal tissues, its role in the OVC needs further study. Verrucous carcinoma and squamous cell carcinoma on 2.KRT 13 protein expression related to clinical or pathological type; 3.KRT 13 protein may serve as a verrucous carcinoma and squamous cell carcinoma prognostic factor prognostic assessment of patients and for oral verrucous cancer gene therapy targets a certain reference value.
