Abstract


SOCS1 silencing the purpose of research dendritic cell-specific anti-tumor mechanism of action, provide a theoretical basis for the clinical application of dendritic cells , and to explore the application prospects of RNA interference in laryngeal cancer gene therapy . Induced cytokines GM-CSF, IL-4 , and TNF-α in vitro amplification of peripheral blood monocyte-derived dendritic cells , characteristic of dendritic cell morphology was observed under an inverted microscope ; build RNA interference vector transfected dendritic shaped cells , Western blot detection SOCS1 expression screening to inhibit the the SOCS1 expression valid target sequence ; flow cytometry dendritic cell surface molecules CD83 , CD86 , HLA -DR expression ; ELISA analysis of IFN-γ in the supernatant content ; MTT method was used to evaluate the dendritic cells to stimulate T cell proliferation and induction of cytotoxic T cell killing activity . The results are dendritic cells induced in vitro cultured successfully under the microscope , the cells were irregularly shaped membrane surface protruding the many branches samples slender projections ; RNA interference vector design by sequencing verification . Interference sequence can significantly cut SOCS1 expression level ; joint of SOCS1 silencing of Hep-2 antigen pulsed dendritic cells can be significantly raised surface marker CD83 ( 85.61 ± 0.96 ) % , CD86 ( 96.86 ± 1.20) % and HLA- DR ( 98.02 ± 0.94 ) % expression ; dendritic cells can effectively stimulate the proliferation of T cells to increase the secretion of IFN-γ , and ultimately enhance the specific cytotoxicity of cytotoxic T cells , effector to target ratio of 50:1 when its cytotoxic activity was significantly higher than that in the control group ( P lt; 0.01 ) . Conclusion the SOCS1 silencing load of Hep-2 antigen dendritic cells can produce efficient and specific anti - the laryngeal cancer immune response .
