Objectives:Carotid body tumor (CBT) is most form of paraganglioma(PGL). The higher incidence of carotid body tumors on plateaus suggests that long-term chronic hypoxia are probably connected with PGL. Otherwise, hyperplasia of paraganglion may result from some chronic diseases accompanied with reduced arterial partial pressure of oxygen(PaO2). We detect hypoxia inducible factor-1α(HIF-1a) and vascular endothelial growth factor(VEGF) in PGL (different pathogenetic locations and malignant grading) by immunohistochemistry and electron microscope analysis. Then we understand the expression of HIF-1a and VEGF in head and neck PGL and the relationship of different locations, properties or clinical grades of PGL and HIF-1a and VEGF.Methods:Thirty-five head and neck PGL with complete clinical and pathological data were investigated after operation. All of them were preliminarily diagnosed at Tianjin Medical University cancer hospital from 2002 to 2010.1. With cases and tumor tissues of patients above, We contrast the expression of HIF-la and VEGF in tumor tissues and control group.2. we analysis the clinical biological characterstics of neck PGL in different clinical indices, such as gender, age, side, tumor features, pathologic diagnosis, local invasion, cervical lymph node metastases, distant metastases and prognosis.we analysis the relevance of the expression of HIF-la and VEGF in neck PGL, the correlation of themselves, and the expression level of HIF-1a and VEGF in different grades, features and hypoxia degree.3. Date was processed with the software of SPSS17.0 to analysis.Results:1. HIF-1αwas expressed in 85.7% of neck PGL, and all cases expressed VEGF. However, neither HIF-la nor VEGF was expressed in the control group.2. The expression level of HIF-1a and VEGF in neck PGL was correlated positively with tumor Shamblin grade(P<0.01).3. The expression level of HIF-1a in neck PGL was correlated positively with VEGF. 4. Age and times with neck PGL was not correlated the expression level of HIF-1a and VEGF(P>0.05).5. The of high area was not correlated the expression level of HIF-1a and VEGF(P>0.05).6. The expression level of HIF-1a and VEGF in malignent neck PGL is high positive.Conclusions:1. It suggests HIF-1a and VEGF play a positive role in the developmental process of head and neck PGL that the expression level of HIF-1aand VEGF was correlated positively with tumor Shamblin grade.2. HIF-1αand VEGF should be related to the occurring of head and neck PGL. HIF-1αmay accelerate angiopoiesis by induced the expression of VEGF and then promote the formation and development of head and neck PGL.3. Age and times with neck PGL was not correlated the expression level of HIF-1a and VEGF.4. The of high area was not correlated the expression level of HIF-1a and VEGF.5. The expression level of HIF-1a and VEGF in malignent neck PGL is high positive.This shows HIF-1a and VEGF may be related to malignant PGL.
