Abstract


Background: With the increase of older people, heart diseases, cerebral increasing incidence, has become one of the most common causes of death. Thrombosis is multifactorial, multi-step pathological process, its mechanism of action include vascular factors, coagulation, anticoagulation and fibrinolysis function abnormalities. Reveal the pathogenesis of thrombosis, the correct clinical diagnosis and treatment of thrombotic diseases have important guiding significance. Growth arrest-specific gene product 6 (growth arrest-specific gene 6, Gas 6) is a vitamin K-dependent protein family members, Axl family, co-ligands. Recently a small number of domestic and foreign literature, Gas 6 plasma levels and gene polymorphism and heart disease risk of thrombosis related to, but mixed results. This study is the first in the Chinese population analysis of plasma levels and Gas 6 gene polymorphism and heart disease related thrombosis and to explore its possible mechanism of action for clinical cored cerebral disease prediction, diagnosis and treatment provide a theoretical basis. Objective: 1. Reveal Gas 6 levels in heart cerebral disease (infarction, acute myocardial infarction) occurred in significance. 2 reveals Gas 6 gene polymorphism and cerebral disease heart (stroke, acute myocardial infarction) correlation. 3 Gas 6 plasma levels and explore the intron c.834 7G gt; A link between genetic polymorphisms and mechanisms. 4. Fill Chinese crowd Gas 6 cerebral disease and cardiac gene polymorphism in the blank. Method: 1. Collect infarction, cerebral hemorrhage, acute myocardial infarction (acute myocardial infarction, AMI) patients and healthy controls blood sample. 2 enzyme-linked immunosorbent assay (Enzyme-Linked ImmunoSorbent Assay, ELISA) were measured in healthy controls and cerebral infarction, cerebral hemorrhage and plasma of patients with AMI Gas 6 levels and high-sensitivity-C reactive protein (high sensitivity C-reactive protein, hs-CRP) levels. 3 in each group were extracted from white blood cells in a blood sample genomic DNA. 4 genomic DNA by polymerase chain reaction (Polymerase Chain Reaction, PCR) amplification and sequencing by gene polymorphisms Gas 6. Results: 1. Cerebral infarction, cerebral hemorrhage and AMI groups Gas 6 plasma levels were significantly higher (P lt; 0.01). (2) cerebral infarction, cerebral hemorrhage and AMI groups Gas 6 introns c.834 7G gt; A Gas 6 of AA genotype was significantly lower than the control group (P lt; 0.01), while the GA genotype Gas 6 levels were significantly higher (P lt; 0.01). 3. Cerebral infarction, cerebral hemorrhage and AMI groups Gas 6 gene c.834 7G gt; A locus of AA genotype and A allele frequency was significantly lower than the control group (P lt; 0.01). 4 cerebral infarction, cerebral hemorrhage and AMI groups plasma hs-CRP levels were significantly higher (P lt; 0.01), Gas 6 levels and hs-CRP level synchronization. 5 infarction, cerebral hemorrhage and AMI associated with smoking, hypertension, high cholesterol and diabetes, these risk factors significantly associated (P lt; 0.01). Conclusions: 1. Gas 6 plasma levels and cerebral infarction, cerebral hemorrhage and AMI-related. 2. Gas 6 gene c.834 7G gt; A locus AA, GG and GA genotype AA genotype Gas 6 plasma levels and cerebral infarction, cerebral hemorrhage and AMI were negatively correlated, suggesting that the level of AA genotype Gas 6 thrombotic diseases may be downregulated by means, thereby reducing the risk of thrombotic diseases. 3. Gas 6 gene c.834 7G gt; A locus genotype AA and A allele frequencies and cerebral infarction, cerebral hemorrhage and AMI was a negative correlation may play a protective infarction, cerebral hemorrhage, AMI role . 4. Gas 6 may be similar with hs-CRP is an acute phase protein in cerebral infarction, cerebral hemorrhage and acute phase of AMI increased reactivity. 5 infarction, cerebral hemorrhage and AMI associated with smoking, hypertension, high cholesterol and diabetes, these risk factors significantly associated.
