Objective: To the study Sinomenine of diabetes (diabetes mellitus, DM) rat kidney expression of transforming growth factor β1 (transforming growth factorβ1, TGF-β1) , to investigate the protective effect and mechanism of sinomenine DM rat kidney . Methods : Healthy male SD rats 45 were randomly divided into 3 groups: normal control group (N ) , DM group (D ) and sinomenine treatment group (S ) , 15 in each group . S group after subcutaneous injection molded sinomenine 20 mg / ( kg · d ) ; D group were given a subcutaneous injection of normal saline administered a total of six weeks . Every two weeks observe the changes in body weight , blood glucose , and 24 h urinary protein excretion of each group were randomly slaughtered three rats , observed by light microscopy of the inferior vena cava blood test serum creatinine (serum creatinine, Scr); renal tissue pathological changes , messenger ribonucleic acid (message ribonucleic acid, mRNA) , TGF-β1 expression in renal tissue was detected by reverse transcription polymerase chain reaction (reverse transcription polymerase chain reaction, RT-PCR) method . Results: With the extension of the observation time , group D rats body mass growth slow , 24 - h urinary protein excretion large , Scr levels high , TGF - β1 mRNA expression levels compared with the N group was statistically significant (P lt ; 0.05). S rats body weight , 24 - h urinary protein excretion and TGF-β1 mRNA expression levels of the D group significantly improved , the difference was statistically significant ( P lt; 0.05 ) ; Scr level and pathological changes of the light microscope has improved , but no significant difference . The correlation analysis showed that renal tissue TGF-β1 mRNA expression levels of 24 - h urinary protein excretion and Scr levels were positively correlated . Conclusion: sinomenine by lowered renal tissue expression levels of TGF-β1 , protective effect on the the DM rat 's kidney .
