Expression of IL-32 in Peripheral Blood of Rheumatism Arthritis Patient and Its Clinical Significance

Abstract


Objective:To detect the expression levels of Interleukin (IL)-32 and Tumor necrosis factor(TNF)-a in peripheral blood of rheumatiod arthritis patients, and analyze the relations between IL-32 and ESR, CRP, RF, DAS28 scores in RA patients. The aim of this study is to identify the clinical significance of IL-32 in RA patient.Methods:We collected 97 specimens of peripheral blood from RA patients, including 36 RA patients with high active phase,33 RA patinets with moderate active phase,28 RA patinets with stable phase,36 non-RA connective tissue diseases(CTD) patients and 30 healthy volunteers. The mRNA levels of IL-32, TNF-a in the peripheral blood mononuclear cells (PBMC) were determind by quantitative real-time PCR (QRT-PCR). The concentration of IL-32, TNF-a in plasma were detected by enzyme linked immunosorbent assay (ELISA), one-way ANOVA and Spearman’s correleation were adopted for statistical analysis.Results:①he mRNA levels of IL-32 and TNF-a in active RA patients were higher than that in healthy control group (P<0.01), and than that in stable RA patients, and than in non-RA CTD patients. There is no significant difference among the stable disease groups, healthy control group and non-RA CTD group in the level of mRNA.②The concentration of IL-32, TNF-a in the plasma of RA patients with active disease was higher than that of healthy control group (P＜0.01).③The expression levels of IL-32 was positively correlated with the level of TNF-a, ESR, CRP, RF, DAS28 scores in the plasma of RA patients (P＜0.01)。Conclusion:①The levels of IL-32 and TNF-a in peripheral blood of active RA patients are higher than those of stable patients and those of control group. The levels of IL-32 in peripheral blood may be used as one reference in order to detect the activity of RA.②The detection of the level of IL-32 in peripheral blood in RA patients can help to understand the development and progress of rheumatiod arthritis and possibly provide one clue for new treatment target for RA.
