Objective:To establish the animal models of streptozotocin induced diabetic nephropathy, and compare the different doses of AKF-PD and PFD, Losartan for the treatment of diabetic nephropathy,Methods:200-240g male Wistar rats were divided into normal group and diabetic group, and the diabetic group were intraperitoneal injected of streptozotocin according to the body weight (50mg/kg); 8 weeks after modeling diabetic, normal rats were divided into 2 groups the normal group and the normal treatment group, while model group rats were divided into 6 groups:model group, AKF-PD 250mg/kg treatment group, AKF-PD 500mg/kg treatment group, AKF-PD 750mg/kg treatment group, PFD 500 mg/kg treatment group, Losartan 20mg/kg treatment group. All rats were weighed and tested glucose per week; after 20 weeks of treatment, collected blood samples to measure of renal function, lefted kidney specimens of HE and PAS staining, and detected renal tissue of collagenⅣand fibronectin by immunohistochemistry.Result:Compared with the normal group, the blood glucose, PAS score, collagen IV and fibronectin protein expression of the model group is increased, while the weight is decreased; Compared with the model group, the weight of AKF-PD in all treatment groups is decreased, and PAS score, collagen IV and fibronectin protein expression are in a dose dependented decrease, while no significant changes in blood glucose and renal function; and compared with the same dose of pirfenidone group, AKF-PD500mg/Kg group decreased the expression of collagenⅣ; and compared with the losartan group, PAS score, collagenⅣ, fibronectin protein expression in AKF-PD 750mg/kg group are decreased.Conclusion:AKF-PD 250,500,750 mg/Kg group of 20 weeks can treat the streptozotocin-induced diabetic rats, it showes a dose-dependence on improving glomerular sclerosis, the 750mg/kg group’s treatment is the best; And the AKF-PD treatment is better than the losartan and the same dose of pirfenidone.
