Therapeutic Effects of BMSCs Combined Puerarin to Treatment Cerebral Ischemia in Rat

ObjectiveBMSCs combined puerarin to treatment cerebral ischemia in rat, to investigate its synergies to promote angiogenesis, and the promotion and mechanism of puerarin to bone marrow stromal cells in vivo.MethodsBMSCs were cultured from the whole bone marrow of rat. The rats were induced to focal cerebral ischemia models with middle cerebral artery occlusion (MCAO). Group of experiments consisted of four groups: MCAO group; BMSCs group (intravenous infusion BMSCs); puerarin group (intravenous injection puerarin), combined group (intravenous infusion BMSCs and puerarin). Then, the four groups were observed at 7th days, 14th days or 28th days after MCAO. BMSCs were labeled by 5-bromodeoxyuridine before transplantation. Neurological functions were evaluated by Zausinger criterion; Volume of cerebral infarction was detected by triphenyltetrazolium chloride staining, immunohistochemistry technologies detected the expression of BrdU and vWF, and immunofluorescence technologies were used to detected the expression of SDF1.Results(1) The NSS scores of combined group, BMSCs group and puerarin group were higher than it of MCAO group (P<0.05); the most highest was the combined group at 28th days, and it has a significant difference from both BMSCs group and puerarin group(P<0.05). (2) The Infarction volume of combined group , BMSCs group and puerarin group were lower than it of MCAO group (P<0.05); the most lowest was the combined group at 28th days, and it has a significant difference from both BMSCs group and puerarin group(P<0.05). (3) The combined group had more BrdU+ cells compared with the BMSCs group(P<0.001). (4) The MVD of combined group , BMSCs group and puerarin group were higher than it of MCAO group(P<0.05); the most highest was the combined group at 28th days, and it has a significant difference from both BMSCs group and puerarin group(P<0.05), but the BMSCs group showed a better trend than the puerarin group (P<0.05). (5) The SDFI expression was increased at the early time of ischemic brain injury, as the time went by, the SDF1 began decreased in the groups of MCAO and BMSCs, the decreased trend of BMSCs group was lower than the MCAO group; but the SDF1 expression had a increased trend in puerarin group and combined group, and combined group was better than the puerarin group (P<0.05).Conclusions(1)Both BMSCs and puerarin can promote angiogenesis, improve neurological functions of MCAO rat, synergistic effect in combination can produce, be helpful for ischemic brain damage repair. (2) SDF1 can be regulated up by puerarin, BMSCs were promoted to migrate to cerebral ischemia area, and to promoted BMSCs treatment cerebral ischemia in rat.
