[ Objective] To observe the inhibitory effect of ginsenoside CK and cisplatin on the proliferation of human lung adenocarcinoma cell line A549 , and a preliminary study of its mechanism of action . Determination of ginsenoside CK cisplatin alone and combined with different drug concentrations ( ginsenoside CK : [ method ] methyl thiazolyl tetrazolium salt (MTT) 50μg/ml, 25μg/ml , 12.5μg/ml , 6.25 μg / ml, 3.125μg/ml, 1.5625μg/ml, DDP: 20μg/ml, 10μg/ml, 5μg/ml, 2.5μg/ml, 1.25μg/ml, 0.625μg/ml), different from the role of time ( 24 h 48h, 72h, 96h) and different order of administration ( administration the give after the ginsenoside CK24h of to DDP, give to ginsenoside CK) and then DDP24h inhibition of A549 cells , using the median-effect principle determines the two drugs morphological changes associated with the effect ; A549 cells were observed under an inverted microscope ; application of flow cytometry for DNA ploidy analysis and TUNEL assay to detect the two drugs alone and their combination with the cell cycle and early apoptotic ; ELISA the assay of cell culture supernatant of VEGF content . [Results ] ginsenoside CK effect showed a concentration and time dependent relationship associated with cisplatin alone ; large doses of the two drugs associated with CI ≈ 1 , additive effect , small dose associated with CI gt ; 1, antagonistic effect ; independent of the inhibitory effect of A549 cells with the administration of the order ( P gt two drugs ; 0.05) ; observed in A549 cells under an inverted microscope morphological changes: significantly reduced the number of cells , cell shrinkage, cell wrinkle shrinkage deformation , shedding floats growth ; flow cytometry A549 cells when the two drugs appear obvious apoptosis peak , S phase , G 2 / M phase cells decreases , cell cycle arrest G 0 / G 1 of the two drugs with apoptosis is higher than with ginsenoside CK or cisplatin ( P lt; 0.05 ) ; two drugs cell culture supernatant of VEGF below with ginsenoside CK or cisplatin group ( P <0.05 ) . [Conclusion ] ginsenoside CK cisplatin large doses associated with inhibitory effects on human lung adenocarcinoma cell line A549 additive effect , small dose associated with antagonistic effects ; big dose associated with significantly induced apoptosis of A549 cells and to reduce the endogenous secretion of VEGF .
