Abstract


Objective: Human immunodeficiency virus (HIV) is the cause of the causative agent of acquired immunodeficiency syndrome (AIDS), in recent years, an increasingly wide range of global HIV epidemic, more and more complex types of virus subtypes. Similarly, China is also facing the increasingly complex genetic diversity of HIV-1. Popular in China, the main subtypes including subtype B, CRFs_BC (including CRF07_BC, CRF08_BC and URFs_BC) and CRF01_AE (they accounted for about 94.9% of all infections). Although these different subtypes share the advantages of different high-risk groups, but almost each of the high-risk groups can be detected all subtypes or popular recombinant this phenomenon for recombination between different subtypes or popular recombinant opportunities. We recently found that gay men (MSM) in Jiangsu Province, subtype B, the the CRF01_AE and CRFs_BC has almost the same proportion of the popular (> 28.6%), which indicates a high possibility of recombination between subtypes or popular recombinant. This paper attempts to looking for new recombinant virus in high-risk groups of Jiangsu Province, and to explore its origins. Method: using nested PCR method for HIV-1 gag-RT, p17, C2-V3 region or full-length genomic sequence and phylogenetic analysis or reorganization. Results: We measured vein related to a group of epidemiologists in Jiangsu Province of drug users (IDU) the HFV-1gag-RT sequence and analysis of its reorganization, the 14 samples, 13 viruses belonging CRF07_BC another one ( named 07JSNJ001) for CRF01_AE and CRF07_BC of recombinant virus. Determination and analysis 07JSNJ001 nearly full-length genome sequence (GenBank accession number FJ238521) showed that the virus is composed of 10 segments spaced CRF07_BC and CRF01_AE fragment of its reorganization map is not the same as the international community have been identified recombinant, a whole new HIV-1 recombinant virus. In addition, through the system of p17 and C2V3 sequence evolutionary analysis, we found that in Wuxi City, Jiangsu Province, a male intravenous drug users infected with HIV-1CRF01_AE (named 07JSWX045). Nearly full-length genome sequence (GenBank accession number FJ441290) Identify and analyze the CRF01_AE virus found: CRF01_AE part (1) .07 JSWX045 genetics Jiangsu Province found CRF01_AE/07_BC reorganization does not have relevance; (2 popular with gay men in Jiangsu Province on the phylogenetic) .07 JSWX045 CRF01_AE virus strains close; (3) of .07 JSWX045 and involved in CRF01/07 recombinant CRF01_AE parental virus occurs in the system and Fujian learn popular CRF01_AE Asian group clustering together; (4). patients JSWX045 did not receive any antiviral therapy, drug analysis found two AZT resistance mutations in HIV-1 reverse transcriptase gene, which is the study of Jiangsu Province the CRF01_AE popular origin and molecular characteristics of the virus strains provide valuable biological information degree point; (5). patients JSWX045 intravenous drug use and heterosexual contact in two high-risk behaviors, suggesting that he might CRF01_AE infected through heterosexual contact, first, and then through the venous drug injection the subtypes spread to other people. Conclusion: This article is the discovery and identification the international first novel recombinant HIV-1 CRF01_AE/07_BC virus, and in the process of looking for the origin of the virus found in Jiangsu Province, an intravenous drug addicts CRF01_AE virus. Independent of the origin of the virus CRF01/07, popular with gay men in Jiangsu Province CRF01_AE closer relationship. The CRF01_AE the virus and CRF01/07 in the CRF01_AE part respectively with subsets of Fujian two CRF01_AE kinship closer. These results indicate that, Jiangsu Province, with the possibility of new high virus and at least two groups of genetic independent CRF01_AE virus is endemic, and may be passed by the Fujian, Jiangsu, indicating a strengthening Jiangsu Province, HIV-1 molecular epidemic learn the necessity of the investigation.
