Abstract


Cervical cancer is one of the most common women's cancer, the incidence ranks second in the world, second only to breast cancer. Of countries with high incidence of cervical cancer, Shanxi Province is one of the areas of high incidence in recent years significantly younger trend, a serious threat to women's health and lives. Cervical intraepithelial neoplasia (CIN), cervical precancerous lesions is also important stage of the prevention and treatment of cervical cancer, the majority of squamous cell carcinoma evolved from precancerous lesions. IL-8 as an important role in inflammation and tumor cells factor in the development of cervical cancer, also plays an important role, the topics to be adopted to varying degrees of cervical lesions by immunohistochemical method study the expression of IL-8 and significance. Objective: Using immunohistochemistry detection method to study the different levels of cervical TCT exfoliated cells, the expression of IL-8, to study cervical exfoliated cells and tissues, IL-8, precisely the extent. Methods: The patient data collected in October 2008 - December 2009 of Obstetrics and Gynecology, the First Affiliated Hospital of Shanxi Medical line TCT check, which TCT positive (ie: TBS grading system lesions in the ASCUS and above ) colposcopy and biopsy for pathological diagnosis. TCT positive samples were taken as the experimental group, 71 cases, cytology results are divided into three groups ASCUS, LSIL, HSIL, TCT examination results for the same period 10 cases of normal specimens randomly selected as a control group; collection TCT positive cervical biopsy tissue 71 cases, divided into inflammation / CIN I, CIN II, of CIN Ⅲ group. Immunohistochemical method detecting cervical cells and cervical biopsy of IL-8 expression were observed with the development of cervical lesions, IL-8 expression and trends, and compare IL-8 in cervical exfoliated cells precisely the extent and tissues. Statistically analyzed using SPSS 16.0 statistical software using x2 test and rank sum test, P lt; 0.05 difference was statistically significant. Results: 1. Collect TCT specimens of 81 cases and preparation of cell wax block cytology knot divided into four groups: normal: 10 cases, ASCUS: 29 Li LSIL: 32 cases, HSIL: 10 cases. Groups of IL-8 expression by immunohistochemical assay, the positive rate 0,31.03%, 62.5%, and 70.0%, respectively. Compare IL-8 expression between normal cells slices with ASCUS, LSIL, HSIL, ASCUS and HSIL positive rate differences were significant sex (P lt; 0.05). Compare IL-8 expression in ASCUS and LSIL, LSIL and HSIL positive differences were not significant (P gt; 0.05). 2.IL-8 ASCUS, LSIL, HSIL positive expression corresponds to the histological diagnosis of CIN II, CIN Ⅲ proportion of 33.33%, 80%, and 100%, respectively. Showed that the IL-8 used in cervical cytology contribute to the detection of high-grade lesions. 3. Collect TCT results corresponding abnormal cervical biopsy paraffin blocks of 71 cases, according to the results of the pathological diagnosis is divided into three groups: inflammation / CIN Ⅰ: 31 cases, CIN Ⅱ: 19 cases, CIN Ⅲ: 21 Li. Groups of IL-8 expression by immunohistochemical assay, the positive rate was 32.26%, 78.95%, 85.71%, respectively. Of IL-8 expression in inflammation / CIN Ⅰ, CIN Ⅱ of CIN Ⅲ positive rate differences were significant sex (P lt; 0.05). Compare IL-8 expression in CIN Ⅱ and CIN Ⅲ positive rate differences were not statistically significant (P gt; 0.05). 4.IL-8 expression in the cell block and tissue blocks comparison: IL-8 in the organization of CIN Ⅱ, CIN Ⅲ expression of the positive rate and cell block LSIL, HSIL expression without significantly with difference (P gt; 0.05) expressed in the organization of inflammation / CIN Ⅰ in positive rate and cell block ASCUS without significantly with difference (P gt; 0.05), showed that IL-8 in the cell block the expression of a certain extent, can reflect IL-8 in tissue expression. Conclusion: 1.TCT prepared cell wax block can be used for screening of cervical lesions. 2.IL-8 expression in normal cells, significantly higher expression in HSIL. 3.IL-8 expression in cell wax block to some extent reflects the organization's expression; positive expression of IL-8 in HSIL, LSIL and more tips for CIN II, CIN III, the positive expression in the ASCUS Tip inflammation / CIN Ⅰ, but its not exclude high-grade lesions.
