Abstract


Purpose to explore topiramate grid column ketone (pioglitazone) nuclear factor of NF - kappa B receptor activation agent with body (receptor activator of NF-κB ligand, RANKL) joint macrophages sets off the stimulus factor (macrophage-colony stimulating factor, M-CSF) induced the osteoclast-like cells (osteoclast-like cells, OLCs) matrix metalloproteinase -9 (matrix metalloproteinase-9, MMP-9) mRNA expression . Separation density gradient centrifugation from human peripheral blood mononuclear cells adherent culture , induced RANKL (50 ng / mL) after joint M-CSF (25 ng / mL) . 14 days after the use of tartrate-resistant acid phosphatase (TRAP) staining and scanning electron microscopy bone slices whether to identify the cells have the characteristics of osteoclasts . After induction success using different concentrations of pioglitazone (0,10-5 ,10-6 ,10-7mol / l) continue to intervene in the culture for 48 hours after the collection of cells in each group , using the reverse transcription polymerase chain reaction ( RT -PCR ) method to detect the expression of MMP-9 mRNA changes . The results ( 1 ) of RANKL joint M-CSF -induced cell morphology generated with characteristics of osteoclasts , tartrate-resistant acid phosphatase (TRAP) staining, with the ability to form resorption pits on bone slices . (2) RT-PCR test results show : human osteoclast-like cells capable of expression of MMP -9 mRNA . (3) in each concentration pioglitazone (10-5 ,10-6 ,10-7mol / l) were cultured for 48 hours after the intervention , MMP- 9 mRNA expression compared with blank control group was significantly decreased ( P lt; 0.01 ) and various concentrations pioglitazone intervention group between MMP-9 mRNA expression was significantly different ( P lt; 0.01 ) . Conclusion RANKL joint induced by M-CSF induced osteoclast-like cells from human peripheral blood . Resulting induction of human osteoclast -like cells capable of expression of MMP -9 mRNA . Pioglitazone can inhibit MMP-9 mRNA expression of osteoclast-like cells , and there is concentration-dependent .
