All the pyemotes (Pyemotes) insects outside of parasitic mites , concealment , gregarious pests potential biological constraints factor . Po mites individual tiny body length is only about 200μm available for classification and identification less morphological characteristics , the classification system is not perfect, especially the lack of an effective distinction between the types of morphological close . In recent years , based on the application of DNA molecular markers provide reference data for the identification of the species . In this study 9 pyemotes populations , each population were randomly selected sample of 3-5 mtDNA-COI gene and ribosomal ITS2 gene was to investigate the species identification pyemotes its phylogenetic through its sequence analysis the molecular evidence for pyemotes classification main conclusions are as follows : 1 , based on mtDNA COI gene sequence analysis showed that the 453bp of COI gene fragments amplified from the 9 pyemotes populations , the average AT content of 70.4% , showing strong AT bias. According to the COI gene among populations evolutionary divergence and phylogenetic tree analysis the ball the abdominal pyemotes group the six populations are divided into three separate species, consistent with the biology and ecology observation results . 2 COI gene sequences of species identification of the 9 pyemotes populations , interspecific evolutionary divergence of 11.9% -17.1 % the intraspecific evolutionary divergence of 0-1.3 % ; through phylogenetic analysis , ball abdominal pyemotes group population 5 (SSXD) phylogenetic relationship with piniperda pyemotes group . 3, using the direct sequencing method on the ribosomal DNA ITS2 zone sequence determination and analysis . Final from 9 pyemotes populations are amplified ITS2 fragment of about 449-491bp , GC content of 32.0% on average . Species determine COI identification of species results were consistent with the kinds of evolutionary divergence between 7.0% -17.8 % the intraspecific evolutionary divergence of 0-0.6 % . 4, COI and ITS2 in species identification Pyemotes play the same role success will Pyemotes the identification to the species level .
