Abstract


Laryngeal carcinoma is one of most common malignant neoplasm in head and neck.In recent years,the morbidity of laryngocarcinoma appears an increasing tendency.It does great harm to people’s health.It has become special focus to investigate the oncogensis,developing, metastasis and management of the tumor from the molecule level. Growth factors and its receptors can induce cells excess generation and canceration and participate some tumorous generation and growth.Platelet-derived growth factor receptor(PDGFR)is single strand hymeno-glucoprotein.PDGFR combined with platelet-derived growth factor(PDGF)can promote kinds of cells multiplicative division and tumor angiogenesis,to participate tumors generate.The blockage of PDGF/PDGFR is a new target for tumor therapy.Objective:To investigate the expression of PDGFR in laryngeal squamous cell carcinoma,and study the correlation between its expression and the clinical-pathological parameters of primary laryngeal squamous cell carcinoma.Method:Expressional level of PDGFR was detected in 36 human laryngeal cancer samples and 18 normal tissues adjacent to cancer respectively,and their correlation with lymph node metastasis and the degree of cell differentiation were studied by immunohistochemistry(IHC) methods.The statistical evaluation was performed with SPSS software for windows.Results:1.The expressions of PDGFR in LSCC are obviously higher than that in normal tissues adjacent to cancer and there was statistical significance (P＜0.01).2.The expressions of PDGFR in well-differentiated samples were lower than moderately and poorly differentiated samples,and there was statistical significance(P＜0.05).3.The expressions of PDGFR in LSCC with lymph node metastasis were obviously higher than that without,and there was statistical significance(P＜0.05).4.The positive rates of PDGFR inⅠ—Ⅱstages andⅢ—Ⅳstages were 41.67%(5/12)and 87.5%(21/24),respectively,there was statistical significance(P＜0.01).5.The positive rates of PDGFR in supra-glottic carcinoma and glttic carcinoma were 63.64%(7/11)and 75%(18/24),respectively.There was no statistical significance(P＞0.05).Conclusions:Our study results suggest that PDGFR are closely related to the malignance degree of laryngeal cancer,the degree of cell differentiation. In the LSCC,PDGFR over expressed,and had relationship with cervical metastasis,which showed PDGFR had a close relationship with LSCC and its metastasis.The study of relationship of PDGFR and laryngeal carcinoma reminds us a new gene therapy of laryngeal carcinoma by inhibiting the function of PDGFR.
