Abstract


Objective : the silicone tube barrier Achilles tendon tendon tissues after injury Achilles tendon intermittent topical application of VEGF, VEGF study the role of endogenous healing in the tendon , to provide a theoretical basis for the clinical application of VEGF treatment of tendon injuries . Methods: 24 healthy rabbits , weighing 2.5- 3.0Kg regardless of male and female , were randomly divided into experimental group and control group , each group after surgery were randomly divided into 2,4,6 weeks three observation phase group , n = 4 only . Achilles tendon right hind limb were taken to establish a tendon injury model : set into the silicone tube ,5 -0 line improvement kessler Select Achilles tendon gastrocnemius bundle branch cut to suture tendons, the date of the experimental group were local added VEGF 100ng after 7,14 21 days administration once again ; corresponding parts of the blank group line tendon suture set into the silicone tube locally without any substance . The 3-0 suture incision , postoperative splint the limb in plantar flexion and knee bit . Postoperative 2,4,6 weeks of three -phase point cut specimens , visual observation of the degree of adhesion , histological staining specimens , image analysis , observation of the damage local area of the fibroblast count as well as the distribution of collagen fibers in the biopsy . Results: 2,4,6 weeks after tendon injury partial local adhesion of visual observation of the experimental and control groups was no significant difference (P> 0.05); 2 weeks after injury localized to fiber cell count experimental group than the control group ( P <0.05), collagen content is also higher than the blank group ( P <0.05 ) ; 4 weeks after injury localized into the fiber cell count experimental group lower than the control group ( P <0.05 ) , but the collagen content higher than the control group (P < 0.05 ) ; six weeks after experimental composition of the fiber cell count lower than the control group ( P <0.05 ) , but the collagen content higher than the blank group ( P <0.05 ) . Longitudinal comparison within each group , the experimental group 2,4,6 weeks after fibroblast cell count decreased (P <0.05), but the increase in collagen fiber content ( P <0.05 ) increase over time ; blank group after 2,4 weeks fibroblast count increases with time increased ( P <0.05 ) , but six weeks fibroblast count is higher than 2,4 weeks ( P <0.05 ) , blank set of collagen fiber content increased with time and increased (P <0.05 ). Conclusion: Topical application of exogenous of VEGF after tendon injury , by promoting damage local angiogenesis and improve local nutrition , and thus to promote and accelerate the proliferation and maturation of fibroblasts , promote secretion of collagen fibers and mature tendon intrinsic healing acceleration.
