Abstract


Objective: To investigate quercetin on wild-type and mutant I502A hkv1.5 role of ion channels and identification of its site of action . Methods: hkv1.5 channel ( wild type and mutant ) gene expression in Xenopus oocytes membrane , using a dual micro- electrode voltage clamp technique both ion channels with quercetin intervention. results: quercetin was voltage , concentration and time-dependent activation of wild-type hkv1.5 channel , while also accelerating the channel activation and inactivation processes . However, the same dose of quercetin did not change mutant I502A ion channel activation and deactivation kinetics properties. In addition , 0.3 mmol / L of hydrogen peroxide activated wild-type hkv1.5 same channel , hydrogen peroxide cells, in the extracellular fluid and then were added two natural antioxidants -100μmol / L quercetin and 4μmol / L tanshinone IIA, tanshinone IIA can find a complete discharge of hydrogen peroxide activation of the channel , and quercetin did not alter the effect of hydrogen peroxide . Conclusions: ( 1 ) activation of the wild -type hkv1.5 quercetin and significantly enhance its ion channel flow ; (2) hkv1.5 S6 ion channel domain mutation residues I502 lift activation of quercetin , the S6 can be assumed domain is an aliphatic neutral amino acid I502 quercetin hkv1.5 channel activating the wild-type locus important role ; ( 3 ) activation of quercetin on the channel is not the classic antioxidant mechanism reflected.
