Abstract


Objective:To study the expression of NF-κB and COX-2 protein in renal cell carcinoma, and to explore their relations with the occurrence, development and metastasis of renal cell carcinoma and the prognosis of patients.Methods:NF-κB and COX-2 protein expression in 48 cases of renal cell carcinoma tissues and 5 cases of normal renal tissues were detected by using immunohistochemical two-step method. The patients were followed up. The relationship between NF-κB, COX-2 protein expression and clinopathological parameters and the prognosis were analyzed.Results:1. The expression of NF-κB and COX-2 protein significantly increased in renal cell carcinoma compared with that in normal renal tissues(P<0.05).2. The expression of NF-κB protein was positively correlated to the clinical stages of renal cell carcinoma(P<0.05), but no correlation between its expression and the pathological grades of renal cell carcinoma (P>0.05). The survival rate of these patients with positive expression of NF-κB protein was significantly lower than that of patients with negative expression of NF-κB protein(P<0.05). The positive expression rate of NF-κB in patients with lymphatic metastasis was higher than that in patients without lymphatic metastasis, but there was no significant difference(P>0.05), and the positive expression rate of NF-κB in patients with distant metastasis was higher than that in patients without distant metastasis, but there was no significant difference(P>0.05). There were no association between the expression of NF-κB protein and tumor diameter of renal cell carcinoma, ages and sexes of patients.3. The expression of COX-2 protein was negatively correlated to the pathological grades of renal cell carcinoma(P<0.05), but no correlation between its expression of NF-κB protein and the clinical stages of renal cell carcinoma(P>0.05). The survival rate of these patients with negative expression of COX-2 protein was significantly lower than that of patients with positive expression of COX-2 protein(P<0.05), but no significantly(P>0.05). There were no association between the expression of COX-2 protein and tumor diameter of renal cell carcinoma, ages and sexes of patients. 4. The Expression of NF-κB and COX-2 protein in renal cell carcinoma had no significantly correlation(P>0.05).Conclusion:1. There may be relationship between the expression of NF-κB and COX-2 protein and the occurrence, progression of renal cell carcinoma.2. The expression of NF-κB may also have association with metastasis of renal cell carcinoma and the prognosis of patients.3. Detecting the expression of NF-κB protein may be useful in diagnosis of renal cell carcinoma and prognosis evaluation of patients.4. Detecting the expression of COX-2 protein may be useful in early diagnosis of renal cell carcinoma.
