Abstract


Objective: To study the traditional Chinese medicine Angelica injection (Angelicasinensis (Oliv) Diels compound injection), ephedrine (Ephedrine), strychnine (Vauqueline), strychnine (Strychnine) multidrug resistance in human leukemia cell line K562 / A02 MDR1 (multidrug resistance 1), MRP (multidrug resistance-associated protein), Topo Ⅱ (topoisomerase Ⅱ), GST-π (glutathione s-transferase) gene and protein expression, and to explore the four traditional Chinese medicine for more than K562/A02 cells reversal of drug resistance pathways. Study subjects: human leukemia resistant cell line: K562/A02 Cell Method: trypan blue staining compound Angelica injection cytotoxicity and MTT (MTT) assay ephedrine, strychnine, disabilities Ning cytotoxicity; using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and immunoblotting (Western Blot) were used to detect the four traditional Chinese medicine in the non-cytotoxic concentration (IC10) under the action K562/A02 cells MDR1, MRP, Topo Ⅱ, GST-πmRNA and protein expression. Results: 1, non-cytotoxic concentration (IC10) of Compound Angelica injection, ephedrine, strychnine effect after, K562/A02 cell MDR1 mRNA and P-gp expression was decreased (P lt; 0.01); while strychnine After the role K562/A02 cells MDR1 mRNA and P-gp expression no significant change (P gt; 0.05). 2, non-cytotoxic concentration (IC10) of the compound effect of Angelica injection after, K562/A02 cell MRP, Topo Ⅱ, GST-πmRNA showed no significant change (P gt; 0.05); Topo Ⅱ protein expression (P lt; 0.01), MRP, GST-π protein had no significant change (P gt; 0.05). Non-cytotoxic concentration (IC10) after strychnine role K562/A02 cells MRP mRNA and protein expression decreased (P lt; 0.01), Topo Ⅱ, GST-πmRNA and no significant changes in protein expression (P gt; 0.05). Non-cytotoxic concentration (IC10) of ephedrine, strychnine effect after K562/A02 cells MRP, Topo Ⅱ, GST-πmRNA and no significant changes in protein expression (P gt; 0.05). Conclusions: 1, non-cytotoxic concentration (IC10) of Compound Angelica injection, ephedrine, strychnine K562/A02 cells can partially reverse drug resistance, due to its effect may be related to decreased cellular MDR1 mRNA in K562/A02 expression, leading to cell membrane expression of P-gp decreased about; rather than cytotoxic concentration (IC10) of strychnine on K562/A02 cell MDR1 mRNA and P-gp expression was not significantly affected. 2, non-cytotoxic concentration (IC10) of Compound Angelica injection can increase the expression of Topo Ⅱ protein partially reversed resistance K562/A02 cells, while Topo Ⅱ mRNA expression levels were not affected; and whose right K562/A02 cells MRP, GST -πmRNA and protein expression was not significantly affected. Non-cytotoxic concentration (IC10) taxi rather through down MRP mRNA and protein expression partially reversed resistance K562/A02 cells, while Topo Ⅱ, GST-πmRNA and protein expression was not significantly affected. Non-cytotoxic concentration (IC10) of ephedrine, strychnine on K562/A02 cell MRP, Topo Ⅱ, GST-πmRNA and protein expression was not significantly affected.
