Phospholipids are composed of a mixture of phosphorus-containing lipid material, the unique bipolar molecular structure gives it many unique physical and chemical characteristics and physiological activity, widely used in food, pharmaceutical and other industries, and various pharmaceutical preparations emulsifier and accessories the ideal raw material. This topic ethanol heat extraction and frozen purified enriched phosphatidylcholine, and with activated carbon to remove pyrogens prepared for injection of soybean lecithin. PC content in the 72-85% and the PE content in the range of 5-10% soybean lecithin product is extracted from the soybean phospholipid. Examine ethanol concentration, solid-liquid ratio, extraction temperature, extraction time, freezing temperature, freezing time on the product yield and PC, PE content. In the case of PC, PE content to meet the requirements to yield the highest distinctions, the results show that the ideal extraction process for ethanol volume fraction of 95%, the extraction of solid to liquid ratio of 1:4, extraction temperature 60 ° C, extraction time 45min, freezing temperature of -15 ℃, freezing time 30min. Response surface analysis on the basis of single-factor test to study solid-liquid ratio, extraction temperature, freezing temperature, freezing time yield and their interactions, optimum process parameters: liquid ratio 1:4.50 extraction temperature 62.75 ℃, freezing temperature of -13.96 ° C, freezing time 30.00min The PC content of 73.89% and 10.00% of the PE content, the yield reached 24.17%. Feasibility of limulus reagent soy lecithin bacteria endotoxin. Interfere with the test results show that 10mg/mL soy lecithin solution is diluted by more than three times, there is no interference of the chromogenic substrate limulus kit quantitative endotoxin content determination. Using the limulus reagent bacterial endotoxin inspection method it is feasible to control the injection grade soybean lecithin bacterial endotoxin. Activated carbon to remove soy lecithin pyrogen process was studied. Examine the amount of active carbon adsorption temperature, adsorption time pyrogen removal. By a single factor test results, with ethanol as the reaction medium, the lowest concentration of endotoxin in 0.5% of the amount of active carbon adsorption temperature 50 ° C, adsorption time 40min conditions, the residues in the sample. By orthogonal test analysis on the basis of single-factor test to obtain the optimal process parameters: 0.6% of the amount of active carbon, heating time 40min, the heating temperature of 50 ° C, the sample bacterial endotoxin concentration 4.41EU / g, the yield was 89.65%. The emulsifying properties of the prepared injection with soybean phospholipid were analyzed, determination of soy lecithin preparation of injectable fat emulsion stability constants, with the stability of the PC content of between 70-85% of the phospholipid prepared intralipid colostrum constant is smaller, and better stability. Having an HLB value between the range of about 7.5-9.5.
