This article click chemistry, ring-opening polymerization and atom transfer radical polymerization combined synthesis has AB3 the asymmetric star, A , 2 (B 3 ) and A of 3 (B 3 ) 3 symmetry hyperbranched star-shaped structure of polylactic acid - polyethylene - methacryloyl block polymers of acryloyloxyethyl phosphorylcholine. Firstly Triassic nitrogen neopentyl alcohol initiator lactide ring-opening polymerization, the synthesis of the azide group of the polylactic acid (PLA-N 3 ), and then through the polylactic acid-containing hydroxy respectively terephthaloyl chloride, trimesoyl chloride esterification reaction, synthetic arms and three arm containing stacked nitrogen group of the polylactic acid (the SPLA-N 3 ). Through butynol bromoisobutylyl bromide esterification reaction with 2 - Synthesis of 2 - methyl - 2 - bromo isobutyrate butynyl alcohol esters, as the initiator by atom transfer radical polymerization synthesis alkynyl group-containing poly acryloyloxyethyl phosphorylcholine (PMPC). Finally, click chemistry synthesis AB3 asymmetric star, A 2 (B 3 ) 2 and A 3 (B 3 ) 3 hyperbranched star polylactic acid - poly 2 - methacryloyloxy ethyl phosphorylcholine block polymer (SPLA- b-PMPC). Structural Characterization of the synthetic intermediates and the final product a 1H-NMR results show that: to terephthaloyl chloride azide-containing end groups of the arms can be obtained a polylactic acid, and the average three-arm product each trimesoyl chloride Nuclear corresponding to 2.85 polylactic acid arm; the AB3, A 2 (B 3 ) 2 and A of 3 (B < sub> 3 ) 3 AB 3 the PLA arm with PMPC the ratio of the number of arms in the polymer as 1:2.93,1:2.87,1:2.51, nearly on the theoretical value of 1:3. Fluorescence spectrophotometric determination of the self-assembly behavior of the different structure of the polymer, the results showed that the synthetic polymer amphiphilic properties, the resulting polymer critical aggregation concentration (CAC) for 0.99 , 1 sup> .76 × 10 -3 sup> mg / mL, A2 (B3) 2 and A3 (B3) 3 hyperbranched star polymer CAC the slightly below AB 3 star polymer. Nanoparticles were solvent evaporation method and dialysis prepared by three structural polymers, transmission electron microscopy and dynamic light scattering test showed that the resulting spherical nanoparticles, particle size 30 2 sup> 00nm. The dialysis method, the resulting nano-particles smaller than the solvent evaporation method; decreases with the increase of the number of polymer arms, the particle size of the nanoparticles. Zeta potential tests show star and hyperbranched polymers Zeta potential of -16.92 - sup> 4.31mV, and with the increase in the number of polymer arms Zeta potential is significantly reduced; compared with polylactic acid 3 (B 3 ) 3 polymer Zeta potential close to 0mV, prove hyperbranched star polymer drug carrier.
