Abstract

Epilepsy is a common disease of neurology. Statistics show that of epilepsy prevalence rate of 7.0 ‰, pursuant to which the estimate of about 900 million people of our existing patients with epilepsy. Epilepsy is not only causing physical harm to the patient, and also seriously affect the quality of life of patients, leading to a series of psychological problems, imposing a heavy burden to families and society. Seizures, cognitive function, behavioral and psychological status, social functioning, self-confidence and a sense of shame, and other factors that may affect the quality of life of patients. Scholars have come to realize the control of seizures is only the comprehensive management of patients with epilepsy as part of social, physiological, psychological and other factors may cause varying degrees of impact on quality of life of patients with epilepsy. The goal of epilepsy treatment from simple to control and reduce the onset of transition to maximize the quality of life of patients. Temporal lobe epilepsy is the most common epilepsy syndrome in adults, accounting for more than 1/3 of the partial epilepsy, more than half of refractory epilepsy. The study confirmed that temporal lobe epilepsy is influenced by genetic factors and environmental factors common role of complex multi-disease susceptibility genes play a role in determining the temporal lobe epilepsy etiology and clinical phenotype. Temporal lobe epilepsy is divided into medial and lateral two clinical subtypes. Mesial temporal lobe epilepsy is the most common drug-refractory epilepsy, one of the early surgical treatment to control seizures and improve cognitive function and improve the prognosis has important significance. Kainic acid (kanic acid, KA) brain excitatory amino acid neurotransmitters - glutamate structural analogues. KA model to simulate human temporal lobe epilepsy ideal conditions: 1. Hippocampus amygdala plays a central role in epilepsy performance; 2. Pathological changes similar to human temporal lobe epilepsy, such as neuronal loss, gliosis and mossy fiber sprouting; like clinical manifestations of human symptomatic temporal lobe epilepsy: after the initial brain damage after a period of asymptomatic quiescent eventually appear recurrent spontaneous seizures; 4. differential role of antiepileptic drugs on KA induced epilepsy. Kainic acid epilepsy model can simulate human temporal lobe epilepsy, is the study of the development and changes of epilepsy, the pathogenic mechanism of effective tools. Kainic acid model has been widely used for the study of epilepsy, many scholars intraperitoneal injection of kainic acid or stereotactic intrahippocampal injection applications successfully induced animal model similar to human cortex epilepsy for the pathogenesis and treatment of epilepsy study provides an important means. The purpose of this experiment is to establish kainic acid induced temporal lobe epilepsy animal model to observe the role of symptoms in the process of its development, EEG, pathological changes and drug treatment, and to explore the pathological mechanism of the model and epileptogenic mechanisms. The first part of the build kainic acid acute kainic acid-induced mouse acute epilepsy model induced epilepsy mouse model study purposes to establish and explore its characteristics. Method were collected from 99 healthy male Kunming mice were randomly divided into saline group (n = 33) and the epileptogenic group (n = 66), epileptogenic intraperitoneal injection of kainic acid 10mg/kg, intraperitoneal injection of saline group physiological brine 35μl / g. Continuous 5h mice were observed after injection the seizures and grading. Intraperitoneal injection of diazepam 4mg/kg give ground when the mice continued seizures up to 1h, control group 3 and epileptogenic group nine simultaneous tracings EEG and brain slices after hematoxylin - eosin staining. change the hippocampus pathological. Results ① behavior learning performance model mice after injection of kainic acid, there may be a wet dog shakes, head and facial muscles clonic limb clonic and generalized tonic-clonic seizures. Saline control group, no seizures. (2) EEG the The status epilepticus mice showed persistent rhythmic spikes, spike and wave or slow wave. ③ pathology studies, bilateral hippocampus may occur neuronal degeneration, CA1 and door area. Conclusion intraperitoneal injection of kainic acid induced epilepsy in mice acute model making simple, seizures incubation period is short, causing epileptic high, and those arising from acute epilepsy model has the same rat model similar to human temporal lobe epilepsy behavior, EEG and neuropathological changes. The second part of penicillin and kainic acid epilepsy model comparison purposes by using penicillin, kainic acid drug production model of epilepsy, and to explore the role of the characteristics and application conditions epileptogenic agent. Experiment 90 healthy male Kunming mice were randomly divided into three groups, the control group (n = 10), penicillin induced epilepsy group (n = 40) and kainic acid the epileptogenic group (n = 40), penicillin epileptogenic by intraperitoneal injection of penicillin 7 × 106 U / kg, the the acid kainic acid induced epilepsy by intraperitoneal injection of the Sea 10 mg / kg, saline group intraperitoneal injection of saline 35 μL, / g. For 5 h after injection, mice were observed for seizures and graded EEG. Results of the control group without seizures, two sets of models seizures, kainic acid induced epilepsy group with penicillin epileptogenic group the Racine graded between 0 ~ Ⅴ Level all levels no significant differences (P gt; 0.05) epileptic behavior of mice Ⅲ ~ V-class incubation period of two sets of epileptogenic group significant difference (t0.05, 78 = 1.990 P lt; 0.05). The low mortality (χ2 = 36.473 P lt; 0.05). Conclusion Intraperitoneal injection of kainic acid-induced animal model is very similar to human temporal lobe epilepsy seizures behavior, pathology, EEG characteristics, is ideal animal model of human temporal lobe epilepsy.
