Investigation of Antitumou Effect on Rabbit Cheek Vx-2 Carcinoma in the Combined Treatment of Laser and 5-FU Sustained-release Dosage

Objective To investigate the photodynamic therapy (PDT) in combination therapy and 5-FU sustained release implant rabbit cheek Vx-2 tumor inhibition and influencing factors, and tried several different detection methods used to study the mechanism. Methods New Zealand rabbits 100 Vx-2 tumor implanted in cheek, the establishment of experimental animal models. Average length until the tumor tissue into 1cm3 randomly divided into blank control group, the laser treatment group ,5-FU sustained release implant group, intravenous injection of 5-FU group ,5-FU sustained release implant and laser combination therapy group of five groups (n = 20). Each group, respectively, corresponding to the laser and 5-FU sustained release implant alone and in combination therapy, two weeks after the weekly measurement of tumor volume, the animals were sacrificed after 5 weeks, the inhibitory rate was calculated, and remove the tumor tissue, observed by HE staining tumor pathology of p53 and PCNA protein expression was detected by immunohistochemical methods and blood concentration detected by high-performance liquid chromatography. Five groups of animal tumors by different treatment methods produce different changes. Inhibition rate of addition to the blank control group, the rest of the group, tumor volume compared with before treatment were not the extent of the decline, including 5-FU sustained-release implants agent and laser tumor volume of the combined treatment group than the other groups was significantly reduced, and calculate the inhibition rate was 93.6 ± 19.5%, statistically analyzed using χ2 test, there are significant differences. (P lt; 0.01) 2.PCNA by immunohistochemical methods in the light microscope estimation results indicate that the animals in each group tumor expression of PCNA in varying degrees, in which 5-FU sustained release implant and laser combination therapy group The expression of PCNA positive rate is low compared with other groups, 10%, statistically analyzed using χ2 test, there are significant differences. (P lt; 0.05). 3.p53 equally by immunohistochemical methods in the light microscope calculation results show that the animals in each group tumor p53 also different levels of expression of p53 expression in the combined treatment group of 5-FU sustained release implant and laser positive rate than other low as 15%, statistically analyzed using χ2 test, there are significant differences. (P lt; 0.05). 4.HE stained blank control group cheek intensive cancer growth, form a complete, circular, polygonal. Sizes, nuclear cytoplasm ratio inversion was malignant change. The rest of the group, the disappearance of most of the nucleolar dense nuclear staining disappeared, membrane integrity, and the emergence of the morphological changes. Especially 5-FU sustained release implant and laser treatment group significantly. 5 HPLC assay blood concentration sustained release agent group and intravenous administration group after the administration, at the initial time within the vein group blood concentration is relatively high, with time, the intravenous administration group 5 -FU through the blood circulation and metabolism, the drug concentration is reduced, while still able to maintain a relatively stable concentration of the drug release agent group. Statistical analysis showed that the two groups there is a very significant difference (P lt; 0.01). Conclusion These results indicate ,5-FU sustained release implant and laser treatment group and the other groups, especially the blank control group, in terms of inhibition of tumor volume, and tumor cell-specific protein expression and cellular morphology changes and aspects there are more obvious differences compared to the release agent group and IV group drug sustained release and stability of the drug concentration, the therapeutic effect of the release agent may be better than the intravenous administration ,5-FU sustained release joint implants and laser treatment options that may exist synergies, better than the other group programs treatment. From the macroscopic shape and size to the change of cell morphology and cell biological characteristics change to the microscopic aspects of the expression of specific proteins are present on tumor growth inhibited. Is recommended.
