Purpose rabbit animal model , connective tissue growth factor (connective tissue growthfactor, CTGF) expression in rabbit conjunctival tissue scarring process , preliminary study CTGF subconjunctival tissue scarring process . Methods 15 New Zealand white rabbits , conjunctiva and subconjunctival tissue of randomly selected an establishment of conjunctival scarring model , the other eye as a control , respectively sacrificed 1,5,9 days after clipping the surgical site . The samples were divided into two parts , a part made ??of biopsy , routine HE staining and immunohistochemistry ; another part for the semi-quantitative reverse transcriptase polymerase chain reaction ( semi -quantitative reverse transcriptionpolymerase chain reaction , RT-PCR ) . Results . Histological observation , one day after visible subconjunctival tissue within the fibroblasts , the increase in the number of inflammatory cells and collagen , and visible blood vessels to dilate ; Tiancheng 5 fiber cells and the number of inflammatory cells after surgery, compared with the first days significantly increased ; the 9th day fibroblasts, decrease in the number of inflammatory cells , collagen increased and compact, and can be seen neovascularization . 2 immunohistochemical staining postoperative day 1 , CTGF positive staining enhanced ; positive staining further enhance postoperative day 5 ; nine days after the positive staining started to diminish , but the group still higher than the control group . Semi-quantitative RT-PCR results show that , one day after CTGFmRNA expression increased postoperative day 5 expression levels continue to increase , to the 9th day expression levels decreased , but still higher than the control group , the difference was significant statistically significant. (p <0.01). Conclusion rabbit conjunctival tissue wound healing process CTGF protein and mRNA expression were increased , suggesting that blocking CTGF gene expression or inhibit its activity , may provide a new therapeutic approach to prevent the failure of glaucoma filtration surgery .
