Abstract


Objective: Objective To comprehend the Cryptosporidium. spp infection state in mammals in Qingdao city. Methods. 388 specimens were collected from dejecture of pigs, cats and dogs in Qingdao Jiaozhou, Jimo and Gaomi and dejecture of experiment animals including rats, mice and rabbits in Parasitology Department of Qingdao University Medical College. Dejecture NS smear directly and iodine-staining, modified acid-fast staining, Auramine-phenol staining and after-staining were used. Images of 825 oocysts were acquied by photograph system. The length, width, perimeter and area of the oocysts were obtained by computer digital image processing system and anlyzed by SPSS software. Crytosporidium. spp egg capsules were separated from dejecture of diarrhea children to infect 2 puppies, and were observed etiologically and pathologically. Result Detection rate of Cryptosporidium. spp in domesticate pig, dog and cat was 4.7%, 22.2% and 0 separately, and detection rate of Cryptosporidium. spp in experimental mouse, rat and rabbit were 33.0% , 40.0% and 15.9% separately. Cryptosporidium. spp detection rate 27.9% in experimental animals were significantly higher than in domesticate animals(7.8%) (X~2=27.96, P<0.0001). Infections puppies show significant diarrhea signs and Cryptosporidium. spp egg capsules were found in dejecture with similar size and shape with human strain.Pathological changes in intestine wall were significant. The avarage length of the oocysts was 3.71μm, raning from 3.68μm to 3.76μm in 95% confidence interval of them. The avarage width was 3.59μm, raning from 3.55μm to 3.63μm in 95% confidence interval of them. The avarage perimeter was 12.54μm and the average area was 10.24μm~2. Conclusion It is first reported in Qingdao city that Cryptospodium. spp infections were defeated in domesticate pig and dog, and experimental animals as rat and rabbit. Cryptospodium. spp human strain has no host specificity in dogs and show significant pathogenicity. For specimen preserved in formalin for long times, 3% hydrochloric acid alcohol decolorised for 5 mins after Auramine-phenol staining, then using modified acid-fast staining can highly raise the detection rate of Cryptosporidium. spp. Data obtained from the computer system are objective and precise,offering scientific foundation for measuring the oocysts and for identifying Cryptosporidium spp.
