Abstract


Within the multiple advantages of this study, based on the realities of the current influenza epidemic status and more than one subtype of highly pathogenic avian influenza virus interspecies transmission , the successful application of bioinformatics theory screened influenza virus epitopes ( influenza virus conservative M the NP proteins CTL epitopes , and H1 , H7 , H9 HA Th cell epitopes ) , design and synthesis of the common for more than one subtype of influenza A virus multi-epitope gene cassette Epi I Epi II. The DNA recombinant pVAX1- to pVAX1 eukaryotic expression plasmid vector was constructed and screened a total of express NP Epi I gene cassette the DNA recombination body pVAX1-NP-Epi I, H5, H3 subtype IV HA Epi II gene co-expression H5HA-Epi II-H3HA1-M2e. Applications BALB / c mice animal models for immunogenicity preliminary study , the the immune results showed that recombinant DNA vaccines induce the production of anti-influenza H1 , H3 , H5 , H7 and H9 HA humoral and cell-mediated immunity , which induced the H5HA , H3HA1 higher antibody levels ; lymphocyte transformation test lymphocyte sub- class the FACS detection and γIFN-ELISPOT cellular immune indicators detect recombinant multi-epitope induced strong cell-mediated immunity , indicating that the recombinant DNA construct good immunogenicity , laid the foundation for the final multivalent vaccines can prevent a variety of subtypes of influenza virus .
