Objective: To observe BSHX method the cultured retinal M (u | ¨) ller cells in hypoxia and TGF-β 2 intervention protective effect and its mechanism of action discussed. Method: 1. Improved enzyme digestion method cultured neonatal SD rat retina M (u | ¨) ller cells. 2 added to the culture medium of sodium dithionite (final concentration of 1.0mmol / L) resulting in hypoxia injury model. Transmission electron microscopy (TEM) observation of retinal M (u | ¨) ller cell ultrastructure. Determination of retinal M (u | ¨) ller cell glutamine synthetase activity and lactate dehydrogenase leakage of enzyme-linked immunosorbent assay. According to [ 3 sup> H] labeled D, L-glutamic acid intake judgment retina M (u | ¨) ller cells of Glu uptake. Results: 1. Retinal M (u | ¨) ller cells under hypoxic conditions ultrastructural pathological damage the BSHX decoction can significantly reduce the pathological damage. Under hypoxic conditions, GS activity in the retina M (u | ¨) ller cells 24h and 48h periods significantly lower than under normal conditions; TGF-β 2 increased hypoxia conditions under the retina M (u | ¨) ller cells reduction of GS activity. BSHX decoction can significantly reduce retinal hypoxia and (or) TGF-β 2 conditions M (u | ¨) ller cells GS activity reduction. Hypoxia retinal M (u | ¨) ller cell viability decreased, TGF-β 2 aggravate hypoxic conditions retinal M (u | ¨) ller reduce cell viability. The BSHX decoction can reduce TGF-β 2 and (or) the simulation under hypoxic conditions retinal M (u | ¨) ller cell viability was reduced. Under hypoxic conditions retinal M (u | ¨) ller cells Glu intake increased, BSHX decoction can increase Glu intake in normal circumstances, but in the TGF-β 2 and hypoxia under the conditions of Glu intake is decreased. Conclusion: 1.BSHX decoction can reduce under hypoxic conditions M (u | ¨) ller organelle damage. Retinal hypoxia and (or) TGF-β 2 conditions M (u | ¨) ller cells GS activity 2.BSHX decoction can improve and increase cell viability, and this may be the prevention and treatment of BSHX method DR one of the means of intervention. 3. Hypoxia and TGF-beta 2 exist under conditions BSHX decoction lowered retinal M (u | ¨) Glu uptake ability of ller cells may be in a single species in the cell environment M (U | ¨) ller cells in a self-protective effect.
