Abstract


Objective: To observe the kidney and promoting blood circulation ( hereinafter referred to as BSHX) Act of cultured retinal Müller cells in the the high sugar TGF -β 2 intervention , and a preliminary study of its mechanism of action for BSHX provide experimental basis for clinical application of the method in the DRP . Method: 1. Improved enzyme digestion cultured neonatal SD rat retinal Müller cells . Transmission electron microscopy (TEM) observation retinal Müller cell ultrastructure . 3 spectrophotometer retinal Müller cells glutamine synthetase (GS) activity and extracellular lactate dehydrogenase (LDH) leakage amount . Β liquid scintillation determination of retinal Müller cells glutamate (Glu) intake . Results: 1. Analog high sugar conditions , retinal Müller cells ultrastructure pathological damage . Herb serum 2.BSHX can improve retinal Müller cells GS activity under the normal conditions of the 12 -hour period ; 12 and a 24 - hour period of TGF- β the GS activity of retinal Müller cells 2 and analog high-sugar co - intervention . 3.BSHX Chinese medicines containing serum can improve retinal Müller cell viability under the normal conditions of the 24-hour period and TGF -β in 2 < / sub> and analog high-sugar co - intervention . The Chinese medicines serum containing 4.BSHX improve the Glu uptake retinal Müller cells in a 24 - hour period under normal conditions ; TGF-β 2 and analog high-sugar intervention under no retinal Müller cells Glu uptake change significantly. Conclusion: The high sugar conditions can cause retinal Müller cells ultrastructure pathological damage , retinal Müller cells in the DRP -time functionality changes . Herb serum 2.BSHX this experiment under different conditions the retinal Müller GS activity can improve the stability of the cell membrane , reducing its permeability , enhanced cell viability , Chinese medicine compound of BSHX prevention DRP 's one of the ways of drug intervention . The 3.BSHX Herb serum TGF-β 2 and high sugar intervention retinal Müller cells Glu uptake had no significant effect , might the BSHX Herb Serum intervention retinal Müller cells . kinds of self-protection role.
