Abstract


The chicken BLB1 and BLB2 copy gene to each other , they are associated with many disease resistance or susceptibility . In order to explore the chicken the BLB gene typing methods and genetic polymorphisms of 14 MHC-B haplotype (59 kb) the BLB1 and BLB2 full-length genomic DNA sequence comparative analysis ; designed two pairs of primers , 6 BWEL-SPF chickens known MHC-B haplotype and Hainan red jungle fowl and the unknown of the Roman chicken MHC-B haplotypes BLB1 and BLB2 the full-length sequence determination confirmed six known single haplotypes and has submitted the corresponding haplotypes exactly ; further using or design of the two pairs of primers , the six known the haplotypes in BLB1 BLB2 the exon 2 were amplified by PCR and by direct sequencing of its preliminary stars , then cloning and sequencing to be verified ; using the design of a pair of primers on the new Hampshire chicken unknown haplotype type BLB1 and BLB2 of the exon 2 PCR amplification, through the direct sequencing their DNA typing , and control the results by five microsatellite DNA markers in the region of the MHC-B genotype determination . According to the above experimental results, the conclusions are as follows : 1) does not exist within the similarity BLB1 and BLB2 of fragment can be specifically amplified to complete BLB1 or BLB2 or wherein any one exon primers . 2 ) must use BLB1 and BLB2 exon 2 sequence in order to correct typing of chicken MHC-B haplotype . 3) in the New Hampshire chicken found three MHC-B haplotype , temporarily named as the new one , better than new and new 3.4 ) based on BLB exon 2 DNA sequences direct contracting type microsatellite markers microsatellite allele , and can be solved the problems of the one-to-many .
