Tongue squamous cell carcinoma (Tongue squamous cell carcinoma) is the most common malignant tumors of the oral and maxillofacial; generally higher degree of malignancy, fast growth, strong infiltration; easy to violations of the tongue, causing the tongue movement is limited, thus affecting eating , swallowing, speech and oral function [1]. The traditional treatment of the disease to take surgical resection, radiotherapy and chemotherapy. Although most of the clinical cases take these means for treatment, but some of the other therapeutic measures are also in-depth research, has to get better results. The growing body of evidence shows that in many influencing factors, angiogenesis is a key factor to affect tumor metastasis and growth. Vascular endothelial growth factor (vascular endothelial growth factor, VEGF) as the most important factor to promote angiogenesis, plays an extremely important role in tumor growth. Thus, scholars try the application VEGF monoclonal antibody to antagonize VEGF to inhibit the growth of malignant tumors, and get a certain effect. VEGF monoclonal antibody carcinoid tumor of the oral and maxillofacial tongue squamous cell carcinoma of the inhibition, the research is less inhibitory effect needs further exploration. This study is based on the status of the above study, VEGF monoclonal antibody in vitro, in vivo two experiments to explore the mechanism of the inhibition of its effect on the tongue squamous cell carcinoma Tca-8113, provided the experimental basis for future clinical application, and to gene and drug treatment of tongue squamous cell carcinoma of the new ideas and theoretical basis. Objective: To investigate VEGF monoclonal antibody Tca-8113 tongue squamous cell carcinoma cells in vitro, and in vivo tumor growth inhibition effect and elucidate its mechanism of action, provided the experimental basis for clinical application. Materials and Methods: In vitro cultured tongue squamous cell carcinoma Tca-8113 cells, divided into a control group and the experimental group; experimental group cells with VEGF monoclonal antibody treatment. Application MTT assay, flow cytometry, Real-time PCR on cell proliferation, cell cycle at different time points, VEGF expression were detected. To confirm that the VEGF monoclonal antibody in vitro inhibition of tongue squamous cell carcinoma Tca-8113, and to elucidate its mechanism of action. Tca-8113 Application of cultured cells to establish nude mice xenograft model (N = 10). The experimental animals were divided into 2 groups: the experimental group applications VEGF monoclonal antibody in tumor injection, the control group received normal saline for 28 days. Detection of the tumor volume at different time points, animals, body weight, and the animals were sacrificed when tumor weight; immunohistochemistry VEGF expression and tumor tissue. VEGF monoclonal antibody in vivo inhibition of tumor growth, as well as VEGF monoclonal antibody generated by antagonizing VEGF expression, reducing tumor blood vessels play a tumor inhibitory effect mechanism further confirmed by the experiments. Results: (1) MTT results show that the experimental group (VEGF monoclonal treatment group) cell proliferation rate was significantly lower than the control group (p lt; 0.01); (2) flow cytometry test results showed that the cell cycle distribution of the experimental group were obvious changes in the experimental group G2 phase cells (7.393 ± 0.323) was significantly more than the control group (2.733 ± 0.055) (p lt; 0.01); (3) Real-time PCR results showed that VEGF mRNA levels of the experimental group was significantly lower than the control group (p lt; 0.01). (4) In vivo experiments showed that the experimental group (VEGF monoclonal treatment group) transplanted tumor volume was significantly lower than the control group (p lt; 0.05) lower than the control group (p lt; 0.01), the final tumor weight, the average gray VEGF expression value was significantly higher (p lt; 0.05). Conclusion: (1) VEGF monoclonal antibody significantly inhibited the in vitro proliferation of tongue squamous cell carcinoma cells, change the cell cycle, down VEGF expression; (2) VEGF monoclonal antibody can significantly inhibit tumor growth in vivo, reducing VEGF in tumor tissue Expression; (3) of VEGF monoclonal antibodies may be via direct inhibition of tumor cells and reduce tumor angiogenesis in vivo to exert the inhibitory effect on the tumor.
