Purpose . Study MN1 (meningioma 1) gene in acute myeloid leukemia (acute myeloid leukemia, AML) expression correlation to explore MN1 expression with clinical data . 2 . Significance of MN1 gene of known cytogenetic and molecular genetics context . Method 1 by fluorescence quantitative PCR (qRT-PCR) method to detect the expression level of MN1 , including 75 cases of AML and 17 cases of remission (CR) in patients with bone marrow or peripheral blood samples and 11 normal controls . The Mann-Whitney U test to compare different grouping MN1 expression level differences . Analysis MN1 expression with age , sex , blood first visit , CR rate and early death (ED) and multivariate analysis was performed using logistic regression . 2 . Detection of NPM1 and FLT3 mutations . The while detect NPM1 A type and B-type mutations using allele - specific PCR MN1 expression in the context of different cytogenetic and molecular genetics , rate comparison with the χ2 test multiple samples using Kroskal- Wallis H rank sum test . Results 1 . Onset , recurrence and CR group MN1 No difference was found in the expression of dynamic observation , but 8 patients , 4 cases of CR patients MN1 expression did not significantly change compared with before induction , including three cases of recurrence within 5-6 months MN1 expression and the onset of the three patients are higher than the the MN1 median expression levels . MN1 expression was higher early mortality ( P = .016) , logistics regression analysis, age (P = .011) and MN1 expression (P = .004) risk factors for early death . 2 allele-specific PCR simultaneously detect NPM1 A -type and B-type mutations , this method is fast , easy and high sensitivity . Not observed differences MN1 in AML risk stratification ; / relapsed AML patients study of the initial issuance of the 75 cases , compared with MN1 high expression group , MN1 low expression of NPM1 / FLT3- related ( P = .009) . AML study of 64 patients with newly reached similar conclusions ( P = .01) . Conclusion 1. MN1 continuing high levels of expression may be a high recurrence rate ; MN1 expression was higher ED rate (P = .016), age confounding factors affect ED exclude the impact of age , still show high MN1 expression of early risk factor for death . Both in the early rounds / relapsed AML or just the initial issuance of the AML , MN1 low expression of NPM1 / FLT3- related .
