Abstract


Gastric cancer is a gastrointestinal cancer, lymph node metastasis is the way of the transfer, an important prognostic factor in the decision. The recent discovery of LYVE-1 and D2-40 is considered to be the specific one of the markers of lymphatic endothelium, and can detect the presence of small amount of lymphatic. By detecting tumor lymphatic density explore the relationship the gastric lymphatic generate with lymph node metastasis, and thus provide the basis for clinical treatment. Objective: To study two new lymphatic tube-specific markers LYVE-1, D2-40 in chronic gastritis, at different stages, different grading gastric tissue expression to explore the relationship between the gastric lymphatic generated with lymph node metastasis. Method: 1. Selected 50 cases of surgical resection of gastric cancer specimens 2007.6 -2007.12 years. Each specimen were confirmed by pathology. Electronic gastroscope biopsy and pathologically confirmed 30 cases of chronic gastritis tissue using RT-PCR assay specimens LYVE-1 gene expression. Application of the immunohistochemical Elivision Act detect LYVE-1, D2-40 expression in gastric tissue analysis of its correlation with the clinical and pathological features of gastric. Results: 1.RT-PCR test results: LYVE-1 positive expression rate of gastric cancer tissues was significantly higher than that in chronic gastritis (P lt; 0.05); poorly differentiated gastric positive rate was significantly higher than high, moderately differentiated gastric cancer (P lt; 0.05); distant metastasis of gastric cancer tissues were significantly higher than in the no distant metastasis (P lt; 0.05). Immunohistochemistry test results: 2.1 LYVE-1 positive product expression in lymphatic endothelial cells, brownish yellow. Gastric carcinoma LYVE-1 positive LVD was significantly higher than that in chronic gastritis (7.01 ± 3.47 / HP vs1.22 ± 1.41 / HP, P lt; 0.05); peritumoral LYVE-1-positive LVD was significantly higher than that of cancer within the organization ( 10.43 ± 3.23 / HP vs4.13 ± 1.55 Ge / HP, P lt; 0.05); Low undifferentiated cancer LYVE-1-positive LVD was significantly higher than the high school differentiation (8.46 ± 1.64 / HP vs5.15 ± 2.95 / HP P lt; 0.05), Ⅲ - Ⅳ period LYVE-1-positive LVD was significantly higher than Ⅰ - Ⅱ (8.53 ± 1.39 / HP vs4.28 ± 2.43 Ge / HP, P lt; 0.05), with lymph node metastasis LYVE- the 1 positive LVD was significantly higher than that without lymph node metastasis (8.53 ± 1.36 / HP vs5.38 ± 2.46 / HP, P lt; 0.05), there LYVE-1-positive LVD distant metastases without distant metastasis was significantly higher than that (9.55 ± 1.21 Ge / HP vs6.28 2.46 Ge / HP, P lt; 0.05). 2.2 D2-40 expression in chronic gastritis and gastric cancer: D2-40 positive expression in lymphatic endothelial cells, brownish yellow. LVD (8.01 ± 4.47 / HP) in gastric cancer tissues was significantly higher than that of chronic gastritis (1.32 ± 1.41 / HP) (P lt; 0.05); peritumoral LVD (11.49 ± 3.53 / HP) was significantly higher than cancer organizations (4.53 ± 1.75 / HP) (P lt; 0.05); the lower undifferentiated carcinoma D2-40 positive LVD was significantly higher than the high school differentiation (9.06 ± 1.64 / HP vs6.15 ± 2.95 / HP, P lt; 0.05) Ⅲ - Ⅳ stage D2-40-positive LVD was significantly higher than stage Ⅰ - Ⅱ (9.06 ± 1.39 / HP vs4.28 ± 2.43 Ge / HP, P lt; 0.05), lymph node metastasis D2-40-positive LVD was significantly higher than that without lymph node metastasis (9.10 ± 1.36 / HP vs7.38 ± 2.46 / HP, P lt; 0.05), have distant metastasis D2-40-positive LVD was significantly higher than that of distant metastasis (10.55 ± 1.21 / HP vs7.38 ± 2.46 pcs / HP, P lt; 0.05). 2.3 D2-40 and LYVE-1-positive LVD comparison: gastric carcinoma and its staging, grading, lymph node metastasis and distant metastasis, as well as chronic gastritis, under the same circumstances, D2-40-positive LVD were higher than LYVE-1-positive LVD (P lt; 0.05). Correlation between the 2.4 D2-40 and LYVE-1 positive LVD: D2-40 and LYVE-1 expression in gastric cancer tissues was significantly positively correlated (r = 0.94, P lt; 0.05). Conclusion: 1. D2-40 and LYVE-1 positive product expression in lymphatic endothelial cells, brownish yellow. Cancer and are mainly located in the week. 2 in gastric cancer tissues LVD with the degree of tumor differentiation, lymph node metastasis and distant metastasis were positively correlated. 3. D2-40 marker superior LYVE-1.
