Abstract


[Objective] To explore the the sunitinib (sunitinib malate) , platelet-derived growth factor BB (PDGF-BB) stimulation of airway smooth muscle cells ( human airway smooth muscle cell HASMCs ) proliferation and migration of its mechanism . [Methods] in vitro cultured human airway smooth muscle cells (HASMCs), divided by the inverted microscope , cell morphology and immunocytochemistry identified : control group , PDGF-BB group , PDGF-BB , with sunitinib intervention group , sunitinib group , tetramethyl the trace base thiazolyl tetrazolium (MTT) colorimetric determination of the effect of PDGF-BB HASMCs proliferation and sunitinib intervention , flow cytometry PDGF-BB and sunitinib on of HASMCs cell cycle modified Boyden microporous membrane the dual slot law observed intervention effect of PDGF-BB HASMCs migration and sunitinib , Western blot detection of of HASMCs the PDGFR-β and AKT phosphorylation . [Results ] ( 1) compared with the control group , PDGF-BB (20ng/ml) significantly induced of HASMCs the proliferation ( P lt; 0.05 ) , sunitinib ( 0.3 9.0nmol / L ) in a concentration -dependent inhibition of PDGF-BB -induced proliferation of HASMCs ; ( 2 ) PDGF -BB ( 20ng/ml) stimulation of HASMCs proportion of S phase of the cell cycle was significantly higher than that in the control group ( P lt; 0.05 ) , the proportion of S phase of the cell cycle of PDGF -BB , with sunitinib intervention group PDGF-BB group (P lt; 0.05); (3) compared with the control group , PDGF-BB (20ng/ml) significantly induced HASMCs migration ( P LT ; 0.05 ) of sunitinib (0.3 9.0nmol / L ) a dose- dependent inhibition of PDGF-BB - induced migration of HASMCs ; (4) PDGF-BB group of HASMCs the PDGFR-β and AKT phosphorylation levels compared with the control group was high ( P lt ; 0.05 ) , PDGF -BB and sunitinib intervention group HASMCs PDGFR-β and AKT phosphorylation levels below the PDGF-BB group . [Conclusion] sunitinib inhibition of PDGF-BB -induced proliferation and migration of HASMCs , by regulating the PI3K/AKT pathway play a role .
