Association of Visfatin Gene Single Nucleotide Polymorphism (-1535C/T) and Plasma Levels with Type 2 Diabetes Mellitus in Ningxia Hui and Han Population

Abstract


Objective To investigate visfatin gene promoter region-1535C / T gene polymorphism and visfatin plasma levels of Ningxia Hui and Han population with type 2 diabetes mellitus. To three generations of immediate living in the Ningxia region and back, back to the Han Chinese type 2 diabetic patients and normal subjects were of Han history of intermarriage as the research object. Back from the Department of Endocrinology, Affiliated Hospital of Ningxia Medical University and the Autonomous Region People's Hospital, collected 102/108 back, in patients with type 2 diabetes mellitus and 101/106 cases no family history of diabetes, Han healthy blood samples by enzyme linked immunosorbent assay (ELISA method) Determination of plasma visfatin level, the application of molecular biology techniques detect visfatin promoter region-1535C / T polymorphism of PCR-RFLP. All subjects were measured blood pressure, body mass index (BMI), waist-hip ratio (WHR) and detection fasting venous plasma glucose (FPG), 2h postprandial blood glucose (2hPG), serum triglyceride (TG), total cholesterol ( TC) and other biochemical indicators. By Case-Control, statistical analysis of the fat gene polymorphism in the Ningxia region back in the population of type 2 diabetes mellitus and normal population distribution characteristics and the gene polymorphism and its plasma levels back to Ningxia Han population with type 2 correlation of diabetes. (1) Hui type 2 diabetes plasma visfatin level between 8.34-48.39ng/ml and average (21.77 ± 8.64) ng / ml, normal control group, plasma visfatin level between 3.59-25.59ng/ml with an average of (13.98 ± 5.50) ng / ml. Plasma visfatin level of the case group and the control group compared to the difference was statistically significant (P lt; 0.05). Case group and control group, BMI, WHR, SBP, FPG, 2hPG, HbA1C, TG, TC, HDL-C compared to the difference was statistically significant (P lt; 0.05). Type 2 diabetes plasma visfatin levels and BMI, WHR, FPG was positively correlated. (2) type 2 diabetes mellitus plasma visfatin level between 6.18-45.27ng/ml and average (18.31 ± 7.4) ng / ml and normal control group, plasma visfatin level between 3.00-25.63ng/ml The average (13.00 ± 6.68) ng / ml. Plasma visfatin level of the case group and the control group compared to the difference was statistically significant (P lt; 0.05). Case group and control group WHR, FPG, 2hPG, HbA1C, TG, TC, HDL-C compared to the difference was statistically significant (P lt; 0.05). Type 2 diabetes plasma visfatin levels and WHR, FPG, 2hPG, HbA1C, TG was positively correlated. (3) visfatin promoter region-1535C / T polymorphism sites three genotypes in the Hui with type 2 diabetes / control group distribution: CC-19/20 (18.6% / 19.8%), CT type 49/49 (48% / 48.5%), the TT type 34/32 (33.4% / 31.7%); T, C allele distribution in both groups: C allele 87/89 (42.6% / 44.1%), T allele 117/113 (57.4% / 55.9%). The distribution of genotype and allele frequencies of polymorphic loci in the two groups was not statistically significant (P gt; 0.05) Another, the the visfatin promoter region-1535C / T genotype and allele frequencies of polymorphic loci three in Hui BMI ≥ 25.0kg/m2 and BMI LT; distribution of 25.0kg/m2 the two groups was not statistically significant (P gt; 0.05) (4) The visfatin promoter region-1535C / T polymorphism sites three genotypes distribution in type 2 diabetes mellitus group / control group: CC-20/25 (18.5% / 23.6%), CT 55/45 (50.9% / 42.4%), the TT type 33/36 (30.6% / 34%); T, C allele distribution in both groups:, C alleles 95/95 (44% / 44.8%), T allele 121/117 (56% / 55.2%). The distribution of genotype and allele frequencies of polymorphic loci in the two groups was not statistically significant (P gt; 0.05) In another, the the visfatin promoter region-1535C / T polymorphism sites three genotypes and allele frequencies in the Han BMI the ≥ 25.0kg/m2 and BMI LT; distribution of 25.0kg/m2 two groups was not statistically significant (P gt; 0.05) (5) In the normal control group and type 2 diabetes, visfatin start the three genotype and allele frequencies of the promoter region-1535C / T polymorphism sites in Ningxia Hui and Han population distribution was not statistically difference (P gt; 0.05). (6) the type 2 diabetic population of the Ningxia Hui TT genotype HDL-C in patients with less than CT and CC genotype (P lt; 0.05) in patients. TT genotype in patients with TG Ningxia, type 2 diabetes mellitus patients compared with CC and CT genotype, the difference was statistically significant (P lt; 0.05). Conclusions (1) Hui type 2 diabetes plasma visfatin, BMI, WHR, SBP, FPG, 2hPG, HbA1C, TG, TC higher than that of the control group, HDL-C than the control group, the difference was statistically significant (P lt; 0.05). Type 2 diabetes plasma visfatin levels and BMI, WHR, FPG was positively correlated. (2) type 2 diabetes mellitus plasma visfatin, WHR, FPG, 2hPG, HbA1C, TG, TC higher than that of the control group, HDL-C than the control group, the difference was statistically significant (P lt; 0.05) Type 2 diabetes plasma visfatin levels and WHR, FPG, 2hPG, HbA1C, TG was positively correlated. (3) with type 2 diabetes and normal control group, the visfatin promoter District-1535C / T polymorphism loci in Ningxia Hui, Han distribution was no significant difference. Visfatin promoter region-1535C / T polymorphism sites Ningxia Hui and Han population with type 2 diabetes was no correlation. Visfatin promoter region-1535C / T polymorphism sites Ningxia Hui, Han population was no correlation between the incidence of obesity. (4) Ningxia Hui with type 2 diabetes population visfatin promoter region-1535C / T polymorphism TT genotype of HDL-C in patients with less than CT and CC genotype patients, in patients with type 2 diabetes mellitus in the TT genotype TG compared with CC and CT genotype. The visfatin promoter region-1535C / T polymorphism TT genotype with abnormal lipid metabolism, Ningxia Hui and Han patients with type 2 diabetes.
