Abstract


Purpose Proteinuria is an important factor in the progress of kidney disease , podocytes protein molecular structure, proteinuria is caused by dysfunction of the important reasons ; renal fibrosis is a common end-stage renal disease pathways , CTGF in the process of renal fibrosis plays a key effect. In rats with adriamycin nephropathy model were established and FSGS minimal change nephropathy model , and mycophenolate mofetil (MMF) to intervene , were used to detect the expression of nephrin and CTGF study of mycophenolate mofetil on the minimal change nephropathy and FSGS therapeutic effect , and to explore the mechanism . Methods 1.SD rats 22 , divided nephropathy model group ( 8 ), MMF treatment group ( 8 ) in the control group ( 6 ) . Model group and treatment group were injected into the tail vein once doxorubicin 7.5mg · kg -1 , injection drug treated rats at day start using MMF20mg · kg -1 · d -1 gavage, other groups equal distilled water for 4 weeks . Control rats injected with saline intravenously once . The groups were injected 1 day before doxorubicin , doxorubicin injection on day 14 after the first 28 days was measured 24-hour urine protein , each in the first 28 days from each of six rats to detect blood biochemistry ; and use immunity histochemistry , western blot were used to detect renal tissue nephrin protein levels . 2.SD rats were divided into control group , adriamycin group , MMF treatment group ( n = 8 rats ) . Nephropathy group , the treatment group were injected into the tail vein once doxorubicin 7.5mg · kg -1 , control rats were injected with saline . Treatment group in the sixth week of MMF20mg · kg -1 · d -1 was suspended in 1ml distilled water , distilled water gavage other groups . Rats in each group were sacrificed 10 weeks 6 ( death due to doxorubicin group 2 ) , the renal pathological changes , and immunohistochemistry , western blot was used to detect the level of CTGF protein in renal tissue .
