Abstract


Deoxyribonucleic acid(DNA) is an important chemical substance in our life process. It is the primary target molecule of most small molecules in organism. The binding formation and site between the small molecules and big molecules influence the physiological and physi-chemical characters of biomolecules. Investigating the mechanism between the small molecules and DNA is thus of vital importance to design the novel drugs and study the pharmacological and toxicity of anticancer drugs.In this paper, the interaction of chromotrope 2R, chromotrope 2B, l-(2-Pyridylazo)-2-naphthol and safranine T, hexakis(imidazole) manganese ([Mn(Im)6](teph)-2H2O) and DNA were studied by electrochemical and spectroscopic (UV/Vis and fluorescence spectra) methods. The appropriate reaction conditions, interaction modes, binding ratios and binding constants between small molecules and DNA have also been determined. The DNA electrochemical sensor has been prepared by using [Mn(Im)6](teph)·2H2O as the electrochemical hybridization label.(1) The electrochemical reactions of the above four dyestuff molecules with DNA on the glassy carbon electrode were studied by the cyclic voltammetry (CV) and differential pulse voltammetry (DPV), while the functionary mechanisms were determined by the UV/Vis spectroscopy and fluorescence spectroscopy. The binding ratios were 2:1 and the binding constants were 5.68 × 105 L2·mol-2, 2.57 ×105 L2·mol-2, 4.56 ×105 L2·mol-2, 3.46 ×105 L2·mol-2
