Clinical examination and biological analysis based on light scattering particles of light emission, light scattering particles labeled microscopic imaging analysis has caused widespread concern, ultra-sensitive immunoassay and DNA probe analysis. Experiments show that application of resonance light scattering (RLS) particles as the mark than the fluorescently labeled sensitivity can be significantly improved. In this paper, on the basis of the ordinary RLS technology, the establishment of a resonance light scattering imaging technology, can be observed in the light scattering properties of the individual aggregate particles, biological macromolecules and drug molecules by light scattering signal detection of single scattering particles, analyzed by HPLC. The study includes the following four aspects: (1) The chromium hydrolysis spectral properties of the polymer, and the use of resonance light scattering (RLS) spectroscopy to study its interaction with nucleic acids. Chromium polymer having a characteristic absorption at 415nm and 580 nm, having a maximum fluorescence emission at 350 nm. Under acidic conditions, chromium polymers with DNA to produce strongly enhanced RLS signal, wherein the peak is located 299.0 nm, whereby discussed the interaction between the inorganic polymeric reagent and the biological macromolecules, and the establishment of the method of analysis of nucleic acids. (2) in an acidic medium, when the presence of a protein, 4 - (p-sulfophenyl) porphyrin (TPPS 4 ) and 4 - (5 - sulfo thienyl) porphyrin [T ( The use of a 488 nm argon ion laser excitation light source of the aggregated particles in the solution, in perpendicular direction with a laser beam, we can observed by an ordinary microscope into a single aggregated particles scattered light, and with a cooled charge-coupled device (CCD) imaging. These digitized image data analysis results show that the protein is proportional to the concentration detected in the focal plane of the number of aggregated particles in the solution, whereby to create a sensitive determination of protein analysis method, and successfully used in the human serum samples Determination of protein.
