Abstract


Objective: rat cardiac gap junction protein Cx43 distribution and content changes and myocardial cells by light microscopy , transmission electron microscope changes in the structure , compared to normal rats and ACOP rat heart of acute carbon monoxide poisoning ( ACOP ) the gap junction the protein Cx43 difference , to determine the pathogenesis of the ACOP rat heart damage and cell Biology basis , to provide a theoretical basis for the ACOP clinical treatment . Methods: 30 young Wistar male rats (3-5 months ) , weight 300g ± 25g . Were randomly divided into experimental group 20 , control group 10 . ( 1 ) the ACOP rat model making : the experimental group were treated with intraperitoneal injection of CO gas , 170ml/kg , preparation of the ACOP animal model , the control group was injected with the same dose of air . 2 hours after blood collection ( tails Law ) and derived the concentration of carboxyhemoglobin in the blood were measured , and ECG . (2) staining : HE staining myocardial cell morphology structural changes ; (3 ) immunohistochemical staining of myocardial cells Cx43 changes in the distribution , computer image analysis to determine the changes in Cx43 content ; ( 4 ) and transmission electron microscopy cardiomyocytes were observed under ultra- changes in the micro- structure , ACOP rats and the difference between the control group . Results : 1 , ACOP rat tail blood content of HBCO concentration was significantly higher with the control group compared to P LT ; 0.05 , significant difference statistically significant . 2 , ACOP rat ECG abnormal heart rhythm changes , with the control group compared to p LT ; 0.01 , the difference was significant ; experimental rats ECG arrhythmia and myocardial ischemia changes , the incidence of the two groups , p The value of LT ; 0.01, significant difference . 3 , HE staining under light microscope was no significant change in the ACOP rat cardiomyocytes structure . Electron microscope, ultrastructure of rat myocardial cells of the experimental group changed obviously . Experimental group : the diffuse myocardial cells mitochondria - severe swelling, crest derangement , part of the crest and film dissolution disappear ; myofibrillar derangement , part of the sarcomere fault disappears ; blurred intercalated disc structure , part of the intercalated disc fusion disappear .
